摘要:
目的研究罗汉果玉竹颗粒对脂多糖(lipopolysaccharide,LPS)诱导的大鼠急性肺损伤(acute lung injury,ALI)模型的保护作用及其可能机制。 方法将58只SPF(specific pathogen free,SPF)级Wistar大鼠按体质量随机分为空白对照组(K组,10只)、模型对照组(M组,12只)、罗汉果玉竹颗粒低剂量组(LY-L组,3.24 g·kg-1,12只)、罗汉果玉竹颗粒高剂量组(LY-H组,6.48 g·kg-1,12只)和泼尼松阳性药物对照组(PN组,5.4 mg·kg-1,12只)。除K组外,其余各组大鼠均经尾静脉注射LPS溶液(6 mg·kg-1)。造模后,各给药组每日灌胃相应药物1次,给药体积为10 mL·kg-1;K组和M组给予等体积蒸馏水。连续给药7天后,腹腔注射10%水合氯醛(3.5 mL·kg-1)麻醉,腹主动脉取血,采用酶联免疫吸附测定(enzyme-linked immunosorbent assay,ELISA)检测血清白细胞介素6(interleukin-6,IL-6)含量;取肺组织,苏木精-伊红(hematoxylin-eosin,HE)染色观察病理变化,免疫组织化学法检测纤连蛋白(fibronectin,FN)、I型胶原(collagen I,COL1A1)及α平滑肌肌动蛋白(α-smooth muscle actin,α-SMA)的表达水平。 结果与K组比较,M组大鼠肺泡结构破坏,肺泡间隔明显增宽,肺泡壁大面积充血、水肿及炎性细胞浸润;血清IL-6含量升高(P<0.01),肺组织FN、COL1A1及α-SMA表达均增加(P<0.01)。与M组比较,LY-L组和LY-H组大鼠血清IL-6含量降低(P<0.01),肺组织病理损伤明显改善,FN、COL1A1及α-SMA表达均降低(P<0.01)。Pearson相关分析显示,血清IL-6含量与肺组织FN、COL1A1及α-SMA表达均呈正相关(P<0.01)。 结论罗汉果玉竹颗粒对LPS诱导的大鼠ALI具有保护作用,其机制可能与减轻肺组织炎症反应、抑制IL-6释放,进而下调肺组织FN、COL1A1及α-SMA的表达有关。
Abstract:
ObjectiveTo study the protective effects of Luohanguo (Siraitiae fructus) Yuzhu (Polygonati odorati rhizoma) granules on LPS-induced ALI in rats and its potential mechanism. MethodsAll 58 SPF-grade Wistar rats were randomized into the blank control group (K group, 10 rats), model control group (M group, 12 rats), low-dose Luohanguo Yuzhu granules group (LY-L group, 3.24 g·kg-1, 12 rats), high-dose Luohanguo Yuzhu granules group (LY-H group, 6.48 g·kg-1, 12 rats) and prednisone positive control group (PN group, 5.4 mg·kg-1, 12 rats) according to body weight. Except for K group, the rats in the remaining groups received LPS solution (6 mg·kg-1) via tail vein injection. After successfully modeling, the treatment groups received the corresponding drugs by gavage once daily, administration volume was 10mL·kg-1; K group and M group were given the same volume of distilled water. After seven consecutive days of administration, the rats were anesthetized with an intraperitoneal injection of 10% chloral hydrate (3.5 mL·kg-1), and blood samples were collected from the abdominal aorta, ELISA was adopted to detect the contents of IL-6; lung tissues were collected for hematoxylin-eosin (HE) staining to observe pathological changes. Immunohistochemistry (IHC) was adopted to measure the expressions of FN, COL1A1 and α-SMA. ResultsCompared with K group, M group observed disrupted alveolar structure, markedly thickened alveolar septa, and extensive congestion, edema, and inflammatory cell infiltration in the alveolar walls; an increase in the contents of IL-6 (P<0.01), the elevated expressions of FN, COL1A1 and α-SMA in lung tissue (P<0.01). Compared with M group, LY-L group and LY-H group demonstrated a decrease in the contents of IL-6 (P<0.01), a significant improvement in lung histopathological injury, the reduction in the expressions of FN, COL1A1 and α-SMA (P<0.01). Pearson correlation analysis showed that the contents of IL-6 were positively related to the expressions of FN, COL1A1 and α-SMA in lung tissue (P<0.01). Conclusion Luohanguo Yuzhu granules have a protective effect against LPS-induced ALI in rats, and its mechanism might be attributed to the attenuation of the inflammatory response in lung tissue, the inhibition of IL-6 release, and the subsequent downregulation of FN, COL1A1, and α-SMA expression in the lungs.
