摘要:
目的分析大黄治疗溃疡性结肠炎的有效活性化合物及作用靶点以探讨其作用机制。 方法应用中药系统药理学技术平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)数据库筛选出大黄中的潜在化学活性物质及相关靶点蛋白,采用基因组注释数据库平台(GeneCards)预测溃疡性结肠炎的作用靶点,并运用UniProt数据库查询与之相对应的基因名称,借助Cytoscape(3.7.2)软件,构建“药物-疾病-靶点”的可视化网络图,通过STRING数据库平台构建蛋白互作网络,再利用Bioconductor平台和R语言进行基因本体(gene ontology,GO)富集分析和京都基因和基因组百科全书(Kyoto encyclopedia of genes and genomes,KEGG)富集分析。 结果通过筛选,共获得大黄的有效化学成分16种,与UC密切相关的关键化学成分共6个,大黄-UC的共同靶点基因25个,获得PPI核心基因分别为MYC、CASP3、JUN、ESR1、PTGS2、CASP8CASP9、IL1B、CCNB1、CDKN1A、CHEK1、PPARG、BAX、NOS2、BCL2;对基因进行富集分析获得43条GO生物学过程和17条KEGG相关信号通路,涉及p53信号通路、多种凋亡通路、铂耐药通路、细胞凋亡通路、细胞周期通路、VEGF信号通路、细胞衰老通路、坏死性通路、ErbB信号通路、MAPK信号通路、NF-κB信号通路、HIF-1信号通路、PI3K-Akt信号通路。 结论大黄可以通过多靶点、多种途径对UC起到治疗作用,为未来进一步进行实验研究奠定了理论基础。
Abstract:
ObjectiveTo analyze the active compounds and the targets of Dahuang (Rhei radix et rhizoma) in the treatment of ulcerative colitis (UC) and explore its mechanism. MethodsTCMSP was applied to screen the potential active chemicals and the relevant target protein of Dahuang, Genecards was used to predict the action targets of UC, UniProt database was used to query the corresponding gene names, network graph visualization of "medicine-disease-target" was constructed by using Cytoscape (3.7.2) software, protein interaction network was built via STRING database, GO enrichment analysis and KEGG enrichment analysis were performed by using Bioconductor platform and R language. ResultsBy screening, 16 active ingredients were obtained from Dahuang, six key chemical ingredients and 25 common target genes of Dahuang and UC were closely related to UC, the obtained PPI core genes were MYC,CASP3, JUN, ESR1, PTGS2, CASP8CASP9,IL1B, CCNB1, CDKN1A, CHEK1,PPARG, BAX, NOS2 and BCL2; a total of 43 GO biological processes and 17 KEGG related signaling pathways were obtained by gene enrichment analysis, involving p53 signaling pathways, many kinds of apoptosis pathways, platinum-resistant pathway, cellular apoptosis pathways, cellular cycle pathways, VEGF signaling pathways, cellular senescence pathways, necrotizing pathway, ErbB signaling pathways, MAPK signaling pathways, NF-κB signaling pathways, HIF-1 signaling pathways and PI3K-Akt signaling pathways. ConclusionDahuang could develop therapeutic effects by many targets and many ways, which could provide a theoretical foundation for further experimental study in the future.
