摘要:
目的探讨活血益气方抗瘤丸(简称KLW)对裸小鼠颅内肿瘤的抑瘤作用及对大鼠的急性毒性。 方法将裸鼠皮下的U87-RFP胶质瘤组织接种于裸鼠颅内,建立裸鼠颅内肿瘤移植模型,按瘤块体积随机分组,分别灌胃KLW高、低剂量,以溶媒为阴性对照,期间测量裸鼠体质量、瘤体荧光面积并观察给药前后裸鼠症状;连续给药26天后处死裸鼠,称量裸鼠瘤质量;选用SD大鼠,按15 mL/kg(体质量),0.44 g生药/mL灌胃给药,每日2次,阴性对照组饮用纯净水。观察给药前后大鼠一般情况、中毒反应及死亡情况,结束观察后解剖大鼠,肉眼观察各组织脏器变化情况。 结果给药期间裸鼠全部存活,体质量正常增长,一般行为学观察未见明显异常。给药26天后KLW低、高剂量组瘤体荧光面积、瘤质量较模型对照组减小。急性毒性试验中给药组所有大鼠在观察期内全部存活,一般行为学观察未见明显异常。KLW组大鼠体质量与阴性对照组比较差异无统计学意义(P>0.05)。剖检大鼠,肉眼观察各组织脏器未见病理改变。 结论KLW低、高剂量对裸鼠颅内移植神经胶质瘤模型的肿瘤生长具有明显抑制作用,大鼠急性实验表明安全性较高。
Abstract:
ObjectiveTo explore the tumor inhibitory effects of blood-activating Qi-invigorating prescription Kangliu Wan (KLW) on intracranial tumors and its acute toxicity test in rats. MethodsIntracranial tumor transplantation models in nude mice were established by inoculating subcutaneous U87-RFP glioma tissue of nude mice into the skull of nude mice, and they were grouped randomly according to the tumor volumes, drenched with high and low doses of KLW respectively, solvent was taken as negative control, to measure body weight, fluorescence area of tumor of the nude mice, to observe the symptoms before and after the medication during the period; to weigh tumor weight of the nude mice when the mice were sacrificed after medicating for 26 days consecutively; SD rats were drenched, 15 mL/kg, 0.44 g crude drug/mL, twice each day, solvent control group was given purified water. To observe general conditions, toxic reaction and the deaths of the mice before and after medication, after observation, to observe the changes of tissues and organs with naked eyes after performing anatomy. ResultsDuring the medication period, all the nude mice survived, their body weight increased normally, and general behavioral observations showed no abnormality. After medicating for 26 days, fluorescence area of tumor and tumor weight of low and high doses groups of KLW lowered compared with model control group. In the acute toxicity, all the rats in the medication group survived during the observation period, general behavioral observations showed no obvious abnormal results. The difference had no statistical meaning when the body weights of the rats in the tested group were compared with these of negative control group (P>0.05). No pathological change has been found in the tissues observed by naked eyes and the rats. ConclusionLow and high dose of KLW possess the notable inhibitory effects on tumor growth in nude mice of intracranial tumor model, and acute toxicity test in rats show higher safety.
