摘要:
目的观察自拟益气活血方对心衰模型大鼠心肌细胞线粒体功能的影响,探讨其治疗心衰的作用机制。 方法将SPF级SD大鼠90只随机抽取15只为正常组,余75只注射阿霉素4周进行造模,将造模成功的大鼠随机分为模型组、依那普利组及益气活血方低、中、高剂量组,每组15只。采用心动超声检测大鼠心功能;采用酶标仪检测心肌细胞内三磷酸腺苷(adenosine triphosphate,ATP)及心肌细胞线粒体内活性氧(reactive oxygen species,ROS)含量;JC-1方法分析心肌细胞线粒体膜电位(mitochondrial membrane potential,MMP)。 结果与模型组比较,依那普利组及益气活血方低、中、高剂量组大鼠左心室射血分数(left ventricular ejection fraction,LVEF)和左心室射血分数缩短(left ventricular fraction shortening,LVFS)均升高(P<0.05),高剂量组和依那普利组左心室舒张末期内径(left ventricular end-diastolic dimension,LVEDD)、左心室收缩末期内径(left ventricular end systolic diameter,LVESD)均缩短(P<0.05)。依那普利组及益气活血方低、中、高剂量组ROS含量较模型组降低(P<0.05),其中益气活血方中剂量组心肌ROS水平降低最明显(P<0.05),而ATP水平和MMP升高(P<0.05),益气活血方高剂量组与依那普利组ATP和MMP比较差异无统计学意义(P>0.05)。 结论益气活血方能改善心衰大鼠细胞线粒体功能,从而对心肌起保护作用,这可能是其调控心室重构和心肌细胞凋亡的重要机制之一。
Abstract:
ObjectiveTo observe the effects of home-made Qi-invigorating blood-activating prescription (QBP) on mitochondrial function of myocardial cells in rat model with heart failure, and to assess its mechanism of treating heart failure (HF). MethodsAmong 90 SPF level SD rats, 15 ones were randomly selected as the normal group, the remaining 75 rats accepted the injection of adriamycin for six weeks, and randomized into the model group, enalapril group, low, moderate and high doses groups of QBP after successfully modeling, with 15 rats in each group. Cardiac ultrasound was used to detect cardiac function in rats, enzyme-labeled instrument was used to detect the contents of adenosine triphosphate (ATP) and reactive oxygen species (ROS) in myocardial cells; JC-1 method was adopted to analyze mitochondrial membrane potential (MMP) in myocardial cells. ResultsCompared with the model group, LVEF and LVFS raised in enalapril group, low, moderate and high doses groups of QBP (P<0.05), the levels of LVEDD and LVESD decreased in high dose group of QBP and enalapril group (P<0.05). The contents of ROS showed a downward tendency when enalapril group, low, moderate and high doses groups of QBP were compared with the model group (P<0.05), among them, the decrease of ROS level was the most notable in moderate dose group of QBP (P<0.05), while the levels of ATP and MMP raised (P<0.05), the difference had no statistical meaning in the levels of ATP and MMP between high dose group of QBP and enalapril group (P>0.05). ConclusionQi-invigorating blood-activating prescription could improve mitochondrial function of myocardial cells, thus protecting cardiac muscle, and this might be one of the important mechanisms of the prescription regulating ventricular remodeling and myocardium cell apoptosis.
