摘要:
目的利用网络药理学方法探讨半夏厚朴汤治疗咳嗽变异性哮喘(cough variant asthma,CVA)的作用机制。 方法通过中药系统药理学数据库与分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)筛选出半夏厚朴汤中各味中药的活性成分及药物靶点;通过GeneCards数据库获取CVA相关基因,将药物靶点与疾病靶点取交集得到半夏厚朴汤治疗CVA的潜在作用靶点;利用Cytoscape软件建立药物-活性成分-CVA靶点网络图;利用STRING数据库和Cytoscape软件构建蛋白-蛋白相互作用网络(protein-protein interactions,PPI);利用DAVID数据库对半夏厚朴汤治疗CVA的作用靶点进行基因本体论功能富集分析(gene ontology,GO)和KEGG富集分析(kyoto encyclopedia of genes and genomes,KEGG)。 结果共筛选得到44个活性成分,97个半夏厚朴汤治疗CVA的潜在作用靶点,富集分析得到GO功能条目141个,KEGG通路97条。 结论半夏厚朴汤可通过多成分、多靶点治疗CVA。
Abstract:
ObjectiveTo discuss the mechanism of Banxia Houpu Tang in the treatment of CVA by utilizing network pharmacology. MethodsThe active ingredients and herbal targets of the herbs in Banxia Houpu Tang were screened out via TCMSP; CVA-related genes were obtained by GeneCards database, the potential targets of Banxia Houpu Tang in the treatment of CVA were gained after getting the intersection of drug targets with disease targets; network plot of herbs-active ingredients-CVA targets was established by utilizing Cytoscape software; protein-protein interactions (PPI) internet was constructed by using STRING database and Cytoscape software; GO and KEGG of the targets of Banxia Houpu Tang in the treatment of CVA were constructed by using DAVID database. ResultsBy screening, 44 active ingredients and 97 potential targets of Banxia Houpu Tang in the treatment of CVA were gained, 141 GO functional entries and 97 KEGG pathways were yielded in enrichment analysis. ConclusionBanxia Houpu Tang could treat CVA via many ingredients and targets.
