摘要:
目的基于网络药理学方法探讨复方斑蝥胶囊治疗非小细胞肺癌(non-small cell lung cancer,NSCLC)的作用机制。 方法从中药系统药理学数据库与分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)筛选复方斑蝥胶囊中11味中药的活性成分及作用靶点,利用药物靶标数据库TTD、人类基因数据库(the human gene database,GeneCards)、美国国家生物技术信息中心(NCBI)筛选NSCLC相关靶点基因,通过STRING数据库对疾病与药物共同靶点构建蛋白-蛋白相互作用(PPI)网络模型,运用Cytoscape 3.5.1软件构建化合物-关键靶点相互作用网络图,利用DAVID6.8数据库对共同靶点进行生物过程和信号通路富集分析。 结果共筛选出复方斑蝥胶囊的210种活性成分和310个作用靶点,与治疗NSCLC相关靶点155个,关键靶点68个,主要通过作用于丝氨酸/苏氨酸特异性蛋白激酶1、肿瘤蛋白质53、血管内皮生长因子A、白细胞介素6、促分裂原活化蛋白激酶3、胱天蛋白酶3、JUN激酶、髓细胞增生原癌基因、肿瘤坏死因子、促分裂原活化蛋白激酶1、促分裂原活化蛋白激酶8、信号转导因子和转录激活因子3(急性期应答因子)、表皮生长因子、前列腺素内过氧化物合成酶2(前列腺素G/H 合成酶和环氧酶)、雌激素受体1关键靶蛋白调控癌症通路、PI3K-Akt信号通路、癌症中的蛋白多糖通路、NSCLC通路等发挥治疗NSCLC的作用。 结论复方斑蝥胶囊可能通过多靶点、多途径治疗NSCLC。
Abstract:
ObjectiveTo explore the mechanism of compound Banmao capsules in the treatment of NSCLC based on network pharmacology method. MethodsThe active ingredients and targets of 11 herbs of compound Banmao capsules were screened from TCMSP, NSCLC-related target genes were screened by utilizing TTD, GeneCards and NCBI, PPI network models of disease-medicine common targets were constructed by adopting STRING database, the interaction network diagram of compound-key targets was constructed by using Cytoscape 3.5.1 software, biological process and signaling pathway enrichment analysis of common targets were carried out by utilizing DAVID6.8 database. ResultsA total of 210 active ingredients and 310 targets were screened from compound Banmao capsules, 155 targets related to NSCLC and 68 key targets, the prescription could treat NSCLC by regulating cancer pathway, PI3K-Akt signaling pathway, proteoglycan pathway in cancer, NSCLC pathway, mainly via acting on AKT1, TP53, VEGFA, IL-6, MAPK3, CASP3, JUN, MYC, TNF, MAPK1, MAPK8, STAT3, EGF, PTGS2 and ESR1 key target proteins. ConclusionCompound Banmao capsules could treatment of NSCLC from many targets and pathways.
