亚麻子大孔树脂纯化物急性毒性实验研究-《西部中医药》

摘要:: 目的：考察亚麻子大孔树脂纯化物（MREF）的急性毒性，为拟定临床用量提供参考。方法：将昆明种小鼠随机分为对照组（NS溶液）和MREF溶液低剂量组(0.000 253 g/mL)、中剂量组(0.002 53 g/mL)、高剂量组(0.025 3 g/mL），每组10只进行实验，根据实验结果测定LD50或进一步实验测定最大给药量。结果：干预7天内对照组，MREF溶液高、中、低剂量组小鼠皮肤、毛发、眼和黏膜、呼吸、中枢神经系统、四肢活动及体质量均无明显变化，无动物死亡；LD50未测出，受试动物灌胃MREF溶液（0.025 3 g/mL）的24小时内最大给药量为33.7 g/kg，相当于成人每天服用MREF 0.12 g/kg(按60 kg体质量计算)的505.5倍，未见毒性反应。结论：低、中、高剂量的MREF溶液与生理盐水对照组比较未出现任何急性毒性反应，由其最大给药量可知该药口服安全，属无毒级。

Abstract:: Objective: To provide the reference of working out clinical dosage by investigating acute toxicity test of macroporous resin extract from flaxseed (MREF). Methods: The mice were randomly allocated to the control group (NS solution), low (0.000 253 g/mL), moderate (0.002 53 g/mL) and high (0.025 3 g/mL) doses groups of MREF solutions, ten mice each group, maximum dosage was detected by further test or LD50 detected according to the results of the test. Results: There was no obvious change in skin, hair, eye and mucous membrane, respiratory system, central nervous system, limb activities and body weight of the mice in the control group, high, moderate and low doses groups of MREF solutions within seven days after intervention, and no animal death; LD50 wasn't detected, maximum dosage of MREF solutions (0.025 3 g/mL) for the tested animals were 33.7g/kg within 24 hours, equivalent to 505.5 times of the adults taking MREF in the concentrations of 0.12 g/kg(calculated by adult body weight 60kg) each day, no toxicity has been seen. Conclusion: No acute toxicity test has been seen when low, moderate and high doses groups of MREF solutions were compared with the control group of physiological saline, the drug is safe for oral administration and of non-toxic grade according to the maximum dose of MREF solution.
