摘要:
目的：观察安坤种子丸对胚泡着床障碍小鼠子宫内膜HOXa-10表达的影响，探讨其对胚泡着床障碍小鼠子宫内膜容受性的影响及其机制。方法：将有规律动情周期的雌性小鼠随机分为正常组、模型组和中药组，每组25只。先期用安坤种子丸对中药组小鼠进行灌胃，雌、雄合笼后观察雌鼠阴栓率，妊娠第4天对中药组和模型组小鼠颈背部皮下注射米非司酮进行干预，制成胚泡着床障碍模型；于妊娠第5、6天处死小鼠,留取子宫标本。采用Q-PCR技术和免疫组化方法检测小鼠子宫内膜HOXa-10基因及蛋白的表达；同时制备HE切片以观察各组小鼠子宫内膜形态结构的变化，统计分析各组间妊娠率、平均着床胚泡数及着床率等指标的变化。结果：安坤种子丸可以提高胚泡着床障碍小鼠子宫内膜HOXa-10基因及蛋白的表达，妊娠率、平均着床胚泡数及着床率等指标虽低于正常组，但高于模型组。结论：安坤种子丸可能通过提高雌鼠子宫内膜上HOXa-10基因及蛋白的表达，一定程度上改善着床障碍小鼠子宫内膜容受性，促进胚泡着床。
Abstract:
Objective: To explpre the influence of AnKun ZhongZi pills on the expressions of endometrium HOXa-10 of the mice with embryo implantation dysfunction, its influence and the mechanism of endometrial receptivity of the mice. Methods: Female mice who present regular oestrous cycles were randomly allocated to the normal group, the model group and herb group, 25 mice each group. The mice in the herb group accepted intragastric administrationof AnKun ZhongZi pills in advance, vaginal plug rates of female mice were observed after the female and male mice were kept in one cage, the mice were intervened by subcutaneous injection of mifepristone at the forth day after the pregnancy to prepare the model with embryo implantation dysfunction; the womb was taken out after the mice were sarcrificed at the fifth and sixth day after the pregnancy. The expressions of endometrium HOXa-10 gene and protein of the mice were detected by Q-PCR technology and immunohistochemical method; HE sections were prepared to observe the changes of morphological structure of the mice in different groups, pregnancy rate, average number of implanted embryos and implantation rate of different groups were statistically analyzed. Results: AnKun ZhongZi pills could improve the expressions of endometrium HOXa-10 gene and protein, pregnancy rate, average number of implanted embryos and implantation rate were lower than these of the normal group, but significantly higher than these of the model group. Conclusion: AnKun ZhongZi pills could improve endometrial receptivity of the mice to a certain degrees and promote embryo implantation by raising the expressions of endometrium HOXa-10 of the mice.
