摘要:
目的：探讨清肝祛湿活血方对非酒精性脂肪肝大鼠肝脏固醇调节元件结合蛋白-1c（Srebp-1c）表达的影响。方法：将雄性SD大鼠随机分为空白组，模型组，多烯磷脂酰胆碱组，清肝祛湿活血方低、中、高剂量组6组。常规饲养7天后，空白组给予普通饲料喂养；其余各组给予高脂饲料喂养+腹腔注射四环素制备非酒精性脂肪肝大鼠模型，共4周。造模成功后清肝祛湿活血方各剂量组用清肝祛湿活血方治疗,多烯磷脂酰胆碱组予易善复治疗，模型组和空白组给予等量蒸馏水。给药4周后观察大鼠血脂、肝组织病理学改变及Western Blot法测定肝脏Srebp-1c表达。结果:与模型组比较,清肝祛湿活血方低、中、高剂量组TG和TC明显降低(P＜0.05)；肝脂肪变性明显减轻；肝脏Srebp-1c表达均明显下降(P＜0.01)。结论:清肝祛湿活血方能下调大鼠肝脏Srebp-1c表达，对非酒精性脂肪肝有较好的治疗作用。
Abstract:
Objective: To explore the effects of QingGan QuShi HuoXue prescription on the expressions of liver sterol regulatory element binding protein-1c (Srebp-1c) of the rats with non alcoholic fatty liver disease (NAFLD). Methods: All SD rats were randomized into the blank group, the model group, phosphatidylcholine group, low, moderate and high doses groups of QingGan QuShi HuoXue prescription. After given conventional feed for seven days, the blank group accepted normal feed; other groups received high fatty feed and intraperitoneal injection of tetracycline to establish the models of NAFLD, for four weeks, different doses groups of QingGan QuShi HuoXue prescription were treated with QingGan QuShi HuoXue prescription, phosphatidylcholine group with rehabilitation therapy, the model group and the blank group the same amount of distilled water after successfully establishing the models. After medicating for four weeks, pathological changes of liver tissue were observed, and the expressions of liver Srebp-1c were detected by Western Blotting method after detecting the blood lipid. Results: Compared with the model group, the levels of TG and TC decreased obviously in low, moderate and high doses groups of QingGan QuShi HuoXue prescription(P＜0.05); liver fatty degeneration alleviated obviously; the expressions of liver Srebp-1c lowered notably(P＜0.01). Conclusion: QingGan QuShi HuoXue prescription could down regulate the expressions of liver Srebp-1c of the rats, which shows better effects in treating NAFLD.
