目的:建立喜马拉雅紫茉莉中葫芦巴碱含量测定的HPLC法,比较种植和野生喜马拉雅紫茉莉中葫芦巴碱含量的差异。方法:采用离子对色谱法,色谱柱:依利特 Sinopak C18(250 mm×4.6 mm,5 μm);流动相:甲醇-冰醋酸-0.05%十二烷基硫酸钠溶液(20∶0.1∶80);流速:1.2 mL/min;检测波长:265 nm;柱温:30 ℃。结果: 葫芦巴碱在 0.2473~1.2360 μg/mL范围内与峰面积线性关系良好,r=0.999 7;葫芦巴碱平均加样回收率为 99.21 %,RSD为0.50%(n=6);甘南产种植和野生喜马拉雅紫茉莉中葫芦巴碱的含量分别为:0.5876、0.3226 mg/g。结论:甘南产种植喜马拉雅紫茉莉中葫芦巴碱的含量高于野生喜马拉雅紫茉莉中葫芦巴碱的含量,可为喜马拉雅紫茉莉的人工种植及推广应用提供理论依据。

Objective: To set up a method for determining the content of trigonelline in Himalayan Mirabilis by HPLC, and compare the differences of the content of trigonelline between wild and cultivated Himalayan mirabilis. Methods: The ion pair chromatography was used, chromatographic column: Elite Sinopak C18 (250 mm×4.6 mm, 5 μm), mobile phase: methanol-glacial acetic acid-0.05% sodium dodecyl sulfate (20:0.1:80), the flow velocity: 1.2 mL/min, detection wavelength: 265nm, column temperature: 30℃. Results: Trigonelline showed good linear relationship in the range from 0. 247 3 to 1.236 0 μg/mL and peak area, r=0.999 7. The average recovery rate was 99.21%, RSD=0.50% (n=6). The contents of trigonelline in cultivated and wild Himalayan mirabilis were 0.587 6 and 0.322 6 mg/g. Conclusion: The contents of trigonelline contained in cultivated Himalayan mirabilis are higher than these contained in wild Himalayan mirabilis, which could provide theoretical evidence for artificial plant, popularization and application of Himalayan mirabilis.
