摘要:
目的：探讨健脾化瘀方对人肝癌HepG-2细胞侵袭、转移及凋亡的影响。方法：选择人肝癌HepG-2细胞作为实验细胞，以CCK-8、细胞迁移实验(Transwell)、流式细胞术分别检测健脾化瘀方对HepG-2细胞的增殖、转移能力以及凋亡的干预作用。结果：药物作用HepG-2细胞48小时的半抑制浓度(IC50)为12.15，72小时的IC50为9.75；药物作用24小时后，HepG-2 细胞迁移率为（42.11±8.49）%；总凋亡率为（20.23±1.15）%。结论：健脾化瘀方对HepG-2细胞有抑制作用，尤以作用48小时的IC50更显著；且对HepG-2细胞有抑制转移和促进凋亡的效应。
Abstract:
Objective: To discuss the effects of spleen-invigorating stasis-eliminating prescription on the invasion, metasis and apoptosis of human liver cancer HepG-2 cells. Methods: Human liver cancer HepG-2 cells were chosen as the experimental cells, the intervention effects of spleen-invigorating stasis-eliminating prescription on the proliferation, metasis of HepG-2 cells were detected by CCK-8, Transwell and flow cytometry. Results: IC50 responses to HepG-2 cells after the medicines acted on for 48 hours was 12.15, IC50 for 72 hours was 9.75; HepG-2 cell mobility was(42.11±8.49)%; total apoptosis rate was (20.23±1.15)%. Conclusion: Spleen-invigorating stasiseliminating prescription possess the inhibitory effects to HepG-2 cells, especially, IC50 after acted on for 48 hours the most significant; and it has the effects of inhibiting metastasis and promoting apoptosis on HepG-2 cells.
