摘要:
目的：通过复习文献筛选三类造模方法，比较各阶段股骨头坏死模型中影像病理学变化，揭示激素性股骨头坏死发生的病理机制。方法：选择同龄健康新西兰大白兔48只为实验动物，雌雄各半，按体质量随机分为4组各12只，即A组(空白组)，B组(单纯激素组)，C组(内毒素加激素组)，D组(马血清加激素组)。用药后第3、6、9周各随机处死2只兔子取双侧股骨头制成标本，并常规组织病理学切片，观察股骨头病理学改变，确定是否造模成功。结果：各组注射相应药物后C、D组9周DR片示股骨头有轻度骨坏死，结合病理切片示造模成功，其中C组股骨头轻度骨坏死出现较早、较明显。部分指标C、D 2组比较差异有统计学意义(P＜0.05)，与B组比较差异有统计学意义(P＜0.05)，与A组比较差异有统计学意义(P＜0.01)。结果：内毒素加激素组和马血清加激素组均成功实现了造模。其中依据病理切片按骨组织形态以内毒素加激素组造模出现坏死征像较早，马血清加激素组次之，单纯激素未获得成功。
Abstract:
Objective: To compare the imaging pathological changes of the models with femoral head necrosis (FHN) at different phases and reveal the pathological mechanism of FHN by screening three modeling methods and reviewing the literature. Methods: All 48 healthy New Zealand rabbits were chosen, half male and half female, randomized into four groups: A group (the blank group), B group (hormone group), C group (endotoxin and hormone group) and D group (horse sera + hormone group) according to body mass. Bilateral femoral heads were taken as the samples after each two rabbits were sacrificed randomly at the third, sixth and ninth week after medicating each day, pathological changes of femoral heads were observed after slicing the conventional tissues to ensure whether the models were prepared successfully. Results: After injecting for nine weeks, DR results of C and D groups showed that femoral heads had slight osteonecrosis, which suggested that the models were set up successfully after combining with pathological slices, among them, slight osteonecrosis appeared earlier and more obviously in C group. The difference had statistical meaning in the comparisons of parts of the indexes between C group and D group (P＜0.05), the difference had statistical meaning when they were compared with these of B group (P＜0.05), the difference had significant statistical meaning when they were compared with these of A group (P＜0.01). Conclusion: Endotoxin+hormone group and horse sera+ hormone group have successfully realized modeling. Necrotic signs appear in bone tissue morphology in endotoxin+hormone group, the next horse sera+ hormone group according to pathological slices, only hormone group hasn’t realized modeling successfully.
