目的:探讨五味子多糖对人胃癌细胞HGC27凋亡的影响及可能的作用机制。方法:采用CCK8实验检测五味子多糖对HGC27细胞的增殖抑制作用,流式细胞术检测五味子多糖对HGC27细胞凋亡的影响,免疫印迹技术检测五味子多糖对HGC27细胞中双特异性磷酸酶4/细胞外信号调节激酶(dual specificity protein phosphatase 4/extracellular signal regulated kinase,DUSP4/ERK)信号通路相关蛋白、增殖及凋亡相关蛋白表达的影响。结果:与正常对照组相比,五味子多糖能明显抑制HGC27细胞增殖,差异具有统计学意义(P<0.05);五味子多糖能显著诱导HGC27细胞的凋亡,差异具有统计学意义(P<0.05);五味子多糖能明显抑制DUSP4、p-ERK、增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)及B淋巴细胞瘤2(B-cell lymphoma-2,Bcl-2)蛋白的表达,且呈浓度依赖性。结论:五味子多糖能明显抑制HGC27细胞的增殖,诱导其凋亡,其作用可能与抑制DUSP4/ERK信号通路活化有关。

Objective: To investigate the impacts of schisandra polysaccharide on human gastric cancer line HGC27 cell apoptosis and its potential mechanisms. Methods: CCK8 assay was carried out to investigate the anti-proliferation effects of schisandra polysaccharide on HGC27 cells, flow cytometry was used to assess the effects of schisandra polysaccharide on the apoptosis of HGC27 cells. immunoblot assay to determine its influence on DUSP4/ERK signaling pathway-related protein, proliferation and apoptosis-related proteins in HGC27 cells. Results: Compared with blank control group, schisandra polysaccharide could obviously inhibit the proliferation of HGC27 cells, and the difference had statistical meaning (P<0.05); it could significantly induce the apoptosis of HGC27 cells, and the difference showed statistical meaning (P<0.05); the results of immunoblot assay showed that it could notably inhibit the expressions of DUSP4, p-ERK, PCNA and Bcl-2 protein, in a concentration dependent manner. Conclusion: Schisandra polysaccharide could notably inhibit the proliferation of HGC27 cells, induce its apoptosis, and its effects might be related to inhibiting DUSP4/ERK signaling pathway activation.
