摘要:
目的：探讨五味子乙素对肾病综合征大鼠的保护作用及其机制。方法：采用尾静脉一次性注射阿霉素(6.5 mg/kg)构建肾病综合征大鼠模型，将大鼠随机分为空白组，模型组及低、中、高剂量五味子乙素组(15、30、60 mg/kg)。测定大鼠肌酐(creatinine，CRE)、尿素氮(urea nitrogen，UREA)、尿量、肌酐清除率、丙二醛(malondialdehyde，MDA)、一氧化氮(nitric oxide，NO)水平及超氧化物歧化酶(superoxide dismutase，SOD)活性，并采用Western Blot方法检测肾组织中核转录因子2(nuclear transcription factor 2，Nrf2)、谷氨酸-半胱氨酸连接酶催化亚基(glutamate L-cysteine ligase catalytic subunit，GCLC)、醌NADH脱氢酶1(quinone NADH dehydrogenase 1，NQO1)及血红素加氧酶1(heme oxygenase 1，HO-1)蛋白表达水平。结果：五味子乙素可改善肾病综合征模型大鼠肾功能，降低CRE、UREA，增加尿量及肌酐清除率，降低血清MDA及NO水平，增强SOD活性及Nrf2转位，并诱导GCLC、NQO1及HO-1蛋白表达，与模型组相比差异有统计学意义(P＜0.05)。结论：五味子乙素能够改善肾病综合征大鼠肾功能，其作用机制可能与活化Nrf2/ARE信号通路有关。
Abstract:
Objective: To observe the protective effects of schisandrin B on rat model with nephrotic syndrome (NS) and study its possible mechanism. Methods: One-off injection of adriamycin from tail vein (6.5 mg/kg) was adopted to establish NS rat models, and the models were randomly allocated to the blank group, the model group, low, moderate and high dose groups of schisandrin B (15, 30 and 60 mg/kg). To detect CRE, UREA, urinary volume, creatinine clearance rate (CCR) and the levels of MDA and NO, and SOD activity, Western blot method was used to measure the levels of Nrf2, GCLC, NQO1 and HO-1 protein expressions. Results: Schisandrin B could improve renal function of NS rats, reduce CRE and UREA, increase urinary volume and CCR, lower the levels of MDA and NO, enhance SOD activity, it could obviously strengthen Nrf2 dislocation, induce the expressions of GCLC, NQO1 and HO-1 protein, and the difference had statistical meaning compared with the model group (P＜0.05). Conclusion: schisandrin B could improve renal function of NS rats, and its mechanism might be related to activating Nrf2/ARE signaling pathway.
