摘要:
目的考察不同提取方法和不同因素水平对猫头刺总黄酮提取率的影响,优选总黄酮提取条件并建立总黄酮含量测定方法。 方法以芹菜素为对照品,以三乙胺为显色剂,采用紫外-可见分光光度法测定猫头刺中总黄酮的含量;单因素实验考察提取方法、浸泡时间、超声时间、溶剂种类和浓度,以及取样量对总黄酮提取率的影响。 结果总黄酮最佳提取条件为取样量0.2 g,加50%乙醇25 mL作为提取溶剂,超声40 min(功率250 W,频率40 KHz)。芹菜素在2.5~15.0 μg/mL浓度范围内与吸光度呈良好线性关系。通过本实验建立的方法测定4批猫头刺总黄酮的含量为2.893~4.860 mg/g,平均回收率104.26%。 结论该研究优选出了猫头刺总黄酮的提取工艺并建立了总黄酮的含量测定方法,为猫头刺的开发和研究提供了可靠依据。
Abstract:
ObjectiveTo survey the effects of different extraction method and the levels of different factors on the extraction rate of total flavonoids from Maotouci (Oxytropis aciphylla Ledeb.), optimize the extraction conditions of total flavonoids and establish a method for the determination of total flavonoids. MethodsWith apigenin as the reference substance and triethylamine as the chromogenic agent, the content of total flavonoids in Maotouci was determined using UV visible spectrophotometry; the effects of extraction method, soaking time, ultrasonic time, solvent type and concentration and sampling amount on the extraction rate of total flavonoids were investigated by single factor experiment. ResultsThe optimum extraction conditions of total flavonoids were sampling amount 0.2 g, adding 50% ethanol, 25 mL, as extraction solvent, ultrasound 40 min (power 250 W, frequency 40 KHz). Apigenin showed a good linear relationship in the range between 2.5 and 15.0 μg/mL and absorbance. Determined by the method in the experiment, the contents of total flavonoids ranged from 2.893 to 4.860 mg/g in four batches of Maotouci, and average recovery rate reached 104.26%. ConclusionThe extraction technology of total flavonoids is optimized and the method for content determination of total flavonoids is established in the study, it could provide the reliable reference for the development and research of Maotouci.
