摘要:
目的观察藏药薰倒牛正丁醇萃取部位对2型糖尿病大鼠血糖、血脂及肝脏总超氧化物歧化酶(total superoxide dismutase,T-SOD)、丙二醛(malonaldehyde,MDA)、谷胱甘肽过氧化氢酶(glutathione peroxidase,GSH-Px)的影响。 方法将60只SPF级SD大鼠随机抽取10只为正常组,其余50只用高脂饲料喂养加腹腔注射1%链脲佐菌素(streptozocin,STZ)溶液(30 mg/kg)建立大鼠糖尿病模型,将造模成功大鼠44只随机分为模型组、阳性对照组(二甲双胍0.09 g/mL)及正丁醇萃取部位高(生药量16.6 g/mL)、中(生药量8.3 g/mL)、低(生药量4.15 g/mL)剂量组。灌胃相应药物或生理盐水1 mL/100 g,每日1次,连续30天。心脏取血处死大鼠,检测总胆固醇(cholesterol,CHOL)、甘油三酯(triglyceride,TG)、γ-羟基丁酸(4-hydroxybutanoic acid,GHB);取心、肝、脾、肾及胸腺称重,计算脏器指数。ELISA法检测肝脏组织匀浆中T-SOD、MDA、GSH-Px含量;取部分肾脏、胰腺,甲醛固定,脱水、包埋、切片、HE染色,做病理学检测。 结果藏药薰倒牛正丁醇萃取部位中、低剂量组能降低糖尿病大鼠血糖及CHOL、TG、GHB、MDA含量,提高GSH-Px含量及超氧化物歧化酶(superoxide dismutase,SOD)活力;中、低剂量组大鼠肾脏、胰腺病理改变与模型组相比有所减轻;各组大鼠各脏器指数比较差异无统计学意义(P>0.05)。 结论藏药薰倒牛正丁醇萃取部位可降低糖尿病大鼠血糖,且对糖尿病大鼠氧化应激有一定减轻作用。
Abstract:
ObjectiveTo observe the effects of n-butanol extract from Tibetan medicine Xundaoniu (Bieber-steinia heterostemon) on blood glucose, blood lipid, the levels of T-SOD, MDA and GSH-Px in liver of the rats with type two diabetes mellitus (T2DM). MethodsTen rats were randomly chosen from 60 SPF SD rats and established as the normal group, the remaining 50 rats accepted high fat feed and intraperitoneal injection of 1% STZ solution (30 mg/kg) to establish T2DM rat models, 44 successful rat models were randomized into the model group, positive control group (metformin 0.09 g/mL), high (crude drug amount 16.6 g/mL), moderate (crude drug amount 8.3 g/mL) and low (crude drug amount 4.15 g/mL) doses groups of N-butanol extract. They were drenched with the corresponding medicine or physiological saline, 1 mL/100 g, once each day, for 30 days consecutively. Rats were killed by taking blood from the heart, to detect CHOL, TG and GHB; the heart, liver, spleen, kidney and thymus were weighed, to calculate organ index. ELISA method was used to measure the contents of T-SOD, MDA and GSH-Px in the homogenate of liver tissue; some kidney and pancreas were taken, fixed with formaldehyde, dehydrated, embedded, sectioned and stained with HE for pathological detection. ResultsModerate and low doses groups of N-butanol extract from Tibetan medicine Xundaoniu could lower the levels of blood glucose, the contents of CHOL, TG, GHB and MDA, raise the contents of GSH-Px and the activity of SOD; pathological changes of the kidney and pancreas in moderate and low doses groups were relieved compared with these of the model group; the difference had no statistical meaning in organ indexes between different groups(P>0.05). ConclusionN-butanol extract from Tibetan medicine Xundaoniu could lower the levels of blood glucose in T2DM rats, and it could relieve oxidative stress to some extent in DM rats.
