摘要:
目的探讨当归多糖对链脲霉素(strep tozotocin,STZ)诱导的糖尿病大鼠糖化血清蛋白(glycated serum protein,GSP)、免疫调节及抗氧化活性的影响。 方法选择SD大鼠10只为空白组,其余经STZ诱导造模成功后,随机分为模型组,吡格列酮组,当归多糖低、中、高剂量组,每组10只。各组给予对应药物灌胃,每日1次,连续灌胃4周。检测各组大鼠糖化血清蛋白、免疫球蛋白水平[免疫球蛋白A(immunoglobulin A,IgA)、免疫球蛋白G(immunoglobin G,IgG)、免疫球蛋白M(immunoglobin M,IgM)]、氧化应激指标[总抗氧化能力(total antioxidant capacity,T-AOC)、超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-PX)]。 结果与模型组比较,当归多糖各剂量组能降低糖尿病大鼠GSP水平(P<0.05),当归多糖各剂量均能不同程度地提高糖尿病血清免疫球蛋白IgA、IgG、IgM含量(P<0.05),当归多糖高、中剂量组均能显著增强糖尿病大鼠血清中SOD活性及T-AOC、GSH-PX含量,并能明显降低MDA、NO含量(P<0.05),当归多糖各剂量组均能增加胰岛素数目,且与浓度呈正相关(P<0.05)。 结论当归多糖能通过减缓自身免疫反应程度和缓解氧化应激来降低糖化血清蛋白,改善胰岛细胞损伤,增加新生胰岛细胞数目,从而达到治疗2型糖尿病的作用。
Abstract:
ObjectiveTo explore the influence of angelica polysaccharides on glycated serum protein (GSP), immune response and oxidative stress in STZ-induced diabetic rats. MethodsTen SD rats were chosen as the blank group, the remaining were randomized into the model group, pioglitazone group, low, moderate and high dosages groups of angelica polysaccharides after successfully modeling induced by STZ, ten rats in each group. Different groups were drenched with the corresponding medicine, once each day, for four weeks consecutively. To detect the levels of GSP, the levels of immunoglobulin (IgA, IgG, IgM), oxidative stress index (T-AOC, SOD, MDA and GSH-PX) of the rats in different groups. ResultsCompared with the model group, different dosage groups of angelica polysaccharides could lower the levels of GSP in diabetic rats (P<0.05), different dosages of angelica polysaccharides could lift the contents of IgA, IgG and IgM to different degrees (P<0.05), high and moderate dosage groups of angelica polysaccharides could notably enhance the activity of SOD in the serum of diabetic rats, the contents of T-AOC and GSH-PX, notably reduce the contents of MDA and NO (P<0.05), different dosage groups of angelica polysaccharides could increase the number of insulin, and positively correlated with concentration (P<0.05). ConclusionAngelica polysaccharides could lower GSP, improve islet cell injury and increase the number of newborn islet cells through relieving the degree of autoimmune response and oxidative stress, thus to treat type two diabetes mellitus.
