摘要:
目的运用网络药理学研究方法,探讨中药栀子治疗IgA肾病(immunoglobulin A nephropathy,IgAN)的潜在靶点及作用机制。 方法使用中药系统药理学数据库与分析平台筛选栀子的有效成分和靶点,通过GeneCards、OMIM、TTD、DrugBank、GAD等数据库查找与IgAN相关的靶点。利用Cytoscape软件分析栀子治疗IgAN的作用靶点与有效成分之间关系网络,通过DAVID数据库分析栀子治疗IgAN的有效靶点的生物学功能和作用机制。 结果根据口服利用度和药物相似性原则筛选出栀子有效成分15个,筛选出的有效成分作用靶点95个。在GeneCards、OMIM、TTD、DrugBank、GAD等数据库中筛选出有关IgAN靶点1346个。通过DAVID数据库进行的基因本体论(gene ontology,GO)和京都基因与基因组百科全书(kyoto encyclopedia of genes and genomes,KEGG)富集分析产生了涉及晚期糖基化终末产物(advanced glycosylation end products,AGEs)-受体(receptor of AGEs,RAGE)信号通路、流体剪切应力与动脉粥样硬化、乙型肝炎、前列腺癌、肿瘤坏死因子(tumor necrosis factor,TNF)信号通路等关键信号通路。 结论栀子通过多靶点、多通路治疗IgAN,其有效作用靶点主要通过炎症因子、血管内皮生长因子、蛋白激酶、细胞凋亡相关因子等作用于多条信号通路,其中AGEs-RAGE信号通路是其发挥治疗作用的重要通路。栀子可能直接或间接参与调节炎症、细胞凋亡等作用机制治疗IgAN。
Abstract:
ObjectiveTo explore the potential targets and mechanism of treating IgA nephropathy (IgA N) with Zhizi (Gardeniae Fructus) using the method of network pharmacology. MethodsTCMSP was used to analyze the active ingredients and targets of Zhizi, GeneCards, OMIM, TTD, DrugBank and GAD databases were utilized to find out the targets related to IgAN. Cytoscape software was applied to analyze the relationship network between the targets and active ingredients of Zhizi in the treatment of IgAN, DAVID database to analyze biological function and mechanism of the effective targets of Zhizi in the treatment of IgAN. ResultsThere were 15 active constituent of Zhizi screened and 95 targets of active ingredients according to the principles of oral availability and drug similarity. 1346 targets related to IgAN were screened from GeneCards, OMIM, TTD, DrugBank and GAD databases. GO and KEGG enrichment analysis performed through DAVID database contained the critical signal pathways such as AGEs-RAGE signal pathway, fluid shear stress and atherosclerosis, hepatitis B, prostate cancer and TNF signal pathway. ConclusionZhizi could treat IgAN through many targets and pathways, its effective targets act on many signal pathways mainly through inflammatory factor, vascular endothelial growth factor, protein kinase, apoptosis-related factors, among which, AGEs-RAGE signal pathway is one of the important pathways of Zhizi playing its therapeutic roles. Zhizi may be directly or indirectly involved in the regulation of inflammation, apoptosis and other mechanisms in the treatment of IgAN.
