摘要:
目的探究丹荔输通汤对慢性输卵管炎性阻塞模型大鼠TLR4/NF-κB信号通路的影响。 方法将84只雌性SD大鼠按随机数字表法抽取11只作为空白组,其余73只大鼠以苯酚明胶化学法构建慢性输卵管炎性阻塞模型,将造模成功大鼠随机分为模型组、西药灌胃对照组、丹荔输通汤中剂量灌胃组及丹荔输通汤高、中、低剂量灌胃+灌肠组共6组,每组11只。模型组灌胃生理盐水;丹荔输通汤高、中、低剂量灌胃+灌肠组分别予14、7、3.5 mg/kg丹荔输通汤灌胃+灌肠;丹荔输通汤中剂量灌胃组予7 mg/kg丹荔输通汤灌胃,西药对照组予10 mg/kg阿奇霉素混悬液灌胃。干预结束后,处死各组大鼠并取输卵管组织,采用苏木精-伊红染色法(hematoxylin-eosin staining,HE)观察各组大鼠输卵管组织形态学变化情况;采用酶联免疫吸附试验(enzyme-linked immunosorbent assay,ELISA)检测各组大鼠血清Toll样受体4(Toll like receptor 4,TLR4)、核因子κB(nuclear factor kappa-B,NF-κB)、肿瘤坏死因子α(tumor necrosis factor α,TNF-α)、白细胞介素6(interleukin-6,IL-6)表达水平。 结果HE结果显示,模型组大鼠输卵管管腔阻塞,绒毛不完整,破坏严重,腔内有炎性渗出,大量炎细胞浸润;各药物组大鼠输卵部组织形态学均较模型组改善,且呈剂量依赖性;与模型组比较,丹荔输通汤各剂量灌胃+灌肠组均能不同程度下调血清TLR4、NF-κB、TNF-α、IL-6表达水平(P<0.01);与西药灌胃对照组比较,丹荔输通汤中、高剂量灌胃+灌肠组血清TLR4、NF-κB、TNF-α、IL-6表达水平明显下调,差异有统计学意义(P<0.01)。 结论丹荔输通汤能够减轻慢性输卵管炎性阻塞模型大鼠输卵管炎性症状,其机制可能与丹荔输通汤调控TLR4/NF-κB信号通路,抑制TNF-α、IL-6等炎性因子表达,从而抑制炎症反应,改善输卵管局部组织炎性状态有关。
Abstract:
ObjectiveTo explore the influence of Danli Shutong Tang on TLR4/NF-κB signaling pathway in rat models with chronic salpingitis-induced blockage. MethodsAll 11 rats were chosen among 84 female SD ones as the blank group according to random number table method, the rest ones were prepared into obstructive models suffering chronic salpingitis using phenol gelatin chemical method, after successfully modeling, rat models were randomized into the model group, the control group (intragastric administration of Western medicine), medium dosage group of Danli Shutong Tang (intragastric administration), high, medium and low dosages groups of Danli Shutong Tang (intragastric administration+enema), altogether six groups, and 11 rats in each group. The model group was drenched with physiological saline; high, medium and low dosages groups of Danli Shutong Tang (intragastric administration+enema) took the decoction via intragastric administration+enema in the concentrations of 14, 7 and 3.5 mg/kg respectively; medium dosage group of Danli Shutong Tang was given Danli Shutong Tang via intragastric administration, 7 mg/kg, the control group of Western medicine (intragastric admini-stration) received 10 mg/kg azithromycin suspension by enteral nutrition. By the end of intervention, the rats of all the groups were sacrificed and the fallopian tubes were taken out, HE staining was adopted to observe the morphology of the fallopian tubes of the rats in different groups; ELISA was applied to detect the levels of TLR4, NF-κB, TNF-α and IL-6 in the rats of all the groups. ResultsHE results displayed that the rats in the model group suffered the obstruction of tuabl lumen, incomplete villi, severe destruction, inflammatory exudation in the lumen and massive inflammatory cell infiltration; the histomorphology of the rats in the groups of medicine was improved compared with the model group, and in a dose-dependent manner; compared with the model group, different dosages groups of Danli Shutong Tang (intragastric administration+enema) could downregulate the expressions of TLR4, NF-κB, TNF-α and IL-6 to varying degrees (P<0.01); compared with the control group (intragastric administration of Western medicine), the levels of TLR4, NF-κB, TNF-α and IL-6 were obviously reduced in moderate and high dosages groups of Danli Shutong Tang (intragastric administration+enema), and the difference had statistical meaning (P<0.01). ConclusionDanli Shutong Tang could alleviate the symptoms, and the decoction could inhibit inflammatory reaction and improve inflammatory state in local tissue of fallopian tube by regulating TLR4/NF-κB signaling pathway and inhibiting the expressions of inflammatory factors such as TNF-α and IL-6.
