摘要:
目的探讨蝉芩颗粒对一氧化氮合酶(inducible nitric oxide synthase,iNOS)/一氧化氮(nitric oxide,NO)-(3′-5′-cyclic guanosine monophosphate,cGMP)-蛋白激酶G(protein kinase G,PKG)通路介导的感染后咳嗽(post infectious cough,PIC)豚鼠神经源性炎症的作用机制。 方法采用H1N1病毒鼻腔滴注染毒模型,将豚鼠随机分为空白组,模型组,蝉芩颗粒高、中、低剂量组,阿斯美组,每组6只。空白组、模型组予生理盐水灌胃;蝉芩颗粒高、中、低剂量组分别以15.20、7.60、3.80 g/(kg·d)剂量蝉芩颗粒灌胃;阿斯美组以25.20 mg/(kg·d)剂量阿斯美灌胃,连续干预14天。干预结束后,采用酶联免疫吸附试验(Enzyme-linked immunosorbent assay,ELISA)测定豚鼠血清中P物质(substance P,SP)、白细胞介素1β(interleukin-1β,IL-1β)、IL-6、肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)、细胞间黏附分子1(intercellular adhesion molecule-1,ICAM-1)表达水平;采用qPCR法检测豚鼠背根神经节中诱导型iNOS、可溶性鸟苷酸环化酶(soluble guanylate cyclase,sGC)、PKG mRNA表达水平;采用蛋白免疫印迹法(western blot,WB)和免疫组织化学法(Immunohistochemistry,IHC)检测豚鼠背根神经节中瞬时受体电位香草酸亚型1(transient receptor potential vanilloid-1,TRPV1)的蛋白表达水平。 结果与空白组比较,模型组豚鼠血清中SP、IL-6、IL-1β、TNF-α、ICAM-1表达水平及背根神经节中iNOS、sGC、PKG mRNA和TRPV1蛋白表达水平均升高(P<0.01);与模型组比较,蝉芩颗粒高剂量组、阿斯美组豚鼠血清SP、IL-6、IL-1β、TNF-α、ICAM-1表达水平及背根神经节中iNOS、sGC、PKG mRNA和TRPV1蛋白表达水平均降低(P<0.05),蝉芩颗粒中剂量组血清SP、IL-6、TNF-α、ICAM-1表达及背根神经节中iNOS mRNA和TRPV1蛋白表达水平降低(P<0.05),蝉芩颗粒低剂量组血清SP和IL-6表达水平及背根神经节中TRPV1蛋白表达水平降低(P<0.05);与阿斯美组比较,蝉芩颗粒高剂量组血清IL-6表达水平升高(P<0.05)、背根神经节TRPV1蛋白表达水平降低(P<0.05),蝉芩颗粒中、低剂量组血清IL-6、IL-1β、背根神经节iNOS、sGC、PKG mRNA表达水平升高(P<0.05),蝉芩颗粒低剂量组血清TNF-α、ICAM-1表达水平升高(P<0.05)。 结论蝉芩颗粒能够通过调控iNOS/NO-cGMP-PKG信号通路,抑制TRPV1通道,减少SP释放,从而减轻PIC豚鼠神经源性炎症。
Abstract:
ObjectiveTo discuss the effects of Chanqin granules on neurogenic inflammation in guinea pigs with post infectious cough (PIC) mediated by iNOS/NO-cGMP-PKG signaling pathway. MethodsAfter establishing the models of H1N1 virus infection by nasal dripping, the guinea pigs were randomized into the blank group, the model group, high, moderate and low doses groups of Chanqin granules, and Asmeton group with six in each group. The blank group and the model group were drenched with physiological saline; high, moderate and low doses groups of Chanqin granules accepted intragastric administration of the medicine at the dose of 15.20, 7.60 and 3.80 g/(kg·d) respectively; and Asmeton group was given compound methoxyphenamine hydrochloride capsules at the dose of 25.20 mg/(kg·d) by gavage, they accepted 14 consecutive days of intervention. By the end of intervention, ELISA was adopted to detect the expressions of SP, IL-1β, IL-6, TNF-α and ICAM-1 in the serum; qPCR method was applied to measure the levels of iNOS, sGC and PKG mRNA in dorsal root ganglion of guinea pigs; the expressions of TRPV1 protein in dorsal root ganglion of guinea pigs were tested using WB and IHC. ResultsCompared with the blank group, the expressions of SP, IL-6, IL-1β, TNF-α and ICAM-1 in the serum were raised, and the levels of iNOS, sGC and PKG mRNA, as well as TRPV1 protein in the model group (P<0.01); compared with the model group, the expressions of SP, IL-6, IL-1β, TNF-α and ICAM-1 in the serum of high dose group of Chanqin granules and Asmeton group were lowered, as well as the levels of iNOS, sGC and PKG mRNA, and TRPV1 protein (P<0.05), the expressions of SP, IL-6, TNF-α and ICAM-1 in the serum and the levels of iNOS mRNA and TRPV1 protein in dorsal root ganglion of guinea pigs were lowered in the moderate dose group of Chanqin granules (P<0.05), the expressions of SP and IL-6 in the serum and the levels of TRPV1 protein in dorsal root ganglion were reduced (P<0.05); Compared with Asmeton group, the expressions of IL-6 were raised (P<0.05) while the expressions of TRPV1 protein in dorsal root ganglion were lowered in high dose group of Chanqin granules (P<0.05), the levels of IL-6 and IL-1β, the expressions of iNOS, sGC and PKG mRNA were increased in moderate and low doses groups of Chanqin granules (P<0.05), the levels of TNF-α and ICAM-1 were elevated in low dose group of Chanqin granules (P<0.05). Conclusion Chanqin granules could inhibit TRPV1 pathway and control the release of SP by regulating iNOS/NO-cGMP-PKG signaling pathway, so as to relieve neurogenic inflammation in PIC guinea pigs.
