目的采用数据挖掘技术,研究《中华人民共和国药典》中含有胆汁类中药成方制剂的用药规律,为胆汁类中药的现代应用和新药开发提供依据。 方法检索《中华人民共和国药典》(2020年版)中含胆汁类中药的中医成方制剂,运用Microsoft Excel、微词云、R语言、Cytoscape等软件,对组成药物进行频次统计和关联规则分析。 结果共纳入87种可口服且含有至少一味胆汁类中药的成方制剂,涉及291味中药。功能主治词云图显示,这些成方制剂主治咽喉肿痛、咳嗽、半身不遂等临床病症。高频药物有牛黄、冰片、甘草、黄芩等。常用药对配伍包括冰片-牛黄、麝香-牛黄、甘草-牛黄、朱砂-牛黄、黄芩-牛黄等。通过关联规则分析构建了中药相互作用网络,识别出4个社团模块,并筛选出一个包含10味中药的核心组方,包括牛黄、冰片、黄连、郁金、水牛角、黄芩、朱砂、栀子、僵蚕、天麻。 结论基于含有胆汁类中药的成方制剂分析发现,其组成以清热药和平肝息风药为主,主要作用于心、肺、肝三脏。核心处方与万氏牛黄清心丸的处方较一致,提示本研究分析的成方制剂与万氏牛黄清心丸在历史沿革和理论渊源上存在关联。
Abstract:
ObjectiveTo provide the basis for modern application and new drug development of bile-containing Chinese patent medicines by studying the medication patterns of the drugs in Pharmacopoeia of the People’s Republic of China using data mining. MethodsWe identified bile-containing Chinese patent medicines from Pharmacopoeia of the People’s Republic of China, and conducted frequency statistics and association rule analysis on constituent herbs using Microsoft Excel, micro word cloud, R language and Cytoscape. ResultsA total of 87 kinds of oral patent medicines containing at least one bile-containing Chinese herbal medicine are included, involving 291 Chinese herbs. The word cloud of therapeutic applications reveals that these classical formula preparations are indicated for treating clinical diseases including pharyngalgia, cough, and hemiplegia. High-frequency drugs are Niuhuang (bovis calculus), Bingpian (borneolum syntheticum), Gancao(Glycyrrhizae radix et rhizoma) and Huangqin (Scutellariae radix). The commonly-used medicinal pairs cover Bingpian-Niuhuang, Shexiang (moschus)-Niuhuang, Gancao-Niuhuang, Zhusha (Cinnabaris)-Niuhuang, and Huangqin-Niuhuang. Association rule analysis was employed to construct and modularize the herbal interaction network, to identify four clusters, and to screen a core prescription containing ten herbs, such as Niuhuang, Bingpian, Huɑngliɑn (Coptidis rhizoma), Yujin (curcumae radix), Shuiniujiɑo (bubali cornu), Huangqin, Zhusha, Zhizi (Gardeniae fructus), Jiɑngcɑn (Bombyx batryticatus) and Tianma (Gastrodiae rhizoma). ConclusionThe analysis of bile-containing Chinese patent medicines demonstrates that heat-clearing herbs and liver-calming wind-extinguishing herbs are their key component classes, mainly acting on three viscera such as heart, lung and liver. The core prescription is more consistent with the prescription of Wanshi Niuhuang Qingxin pills, suggesting the historical evolution and theoretical origin of the prepared herbal medicines under investigation in this research and Wanshi Niuhuang Qingxin pills.
