摘要:
运用系统药理学和生物信息学方法,预测并探究国医大师张磊涤浊法经验方治疗肺癌的关键靶点及作用机制。 方法通过中药系统药理学数据库与分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)数据库筛选涤浊法经验方的有效化合物及作用靶点;在GeneCards数据库获取肺癌相关靶点,与药物作用靶点映射后得到交集作用靶点;利用DAVID数据库进行基因功能(Gene ontology,GO)及京都基因与基因组数据库(Kyoto encyclopedia of genes and genomes,KEGG)通路富集分析;借助TCGA数据库进行差异表达分析,并将差异基因与有效化合物进行分子对接。 结果筛选获得涤浊法经验方有效化合物207个、作用靶点162个;肺癌相关疾病靶点23 701个,映射后得到核心作用靶点51个。DAVID数据库富集分析显示,涤浊法经验方可能通过调控凋亡过程、细胞增殖、基因表达等生物学过程,及TNF信号通路、HIF-1信号通路、PI3K-Akt信号通路等途径发挥抗肺癌作用。“中药-化合物-靶点-通路”综合网络分析表明,槲皮素、木犀草素、山柰酚、汉黄芩素及甘草查耳酮A为作用范围较广的关键有效化合物。差异分析结果显示,相较于低表达者,BCL2、COL1A1、TNFRSF1A基因高表达患者的总体生存率更低(P<0.05);且BCL2、COL1A1在正常组织与肺癌组织中的表达差异具有统计学意义(P<0.05);关键靶点BCL2、COL1A1与关键有效化合物的结合活性良好。 结论初步揭示了涤浊法经验方治疗肺癌的分子机制,挖掘出BCL2、COL1A1有望作为治疗肺癌的关键靶点,为后续深入研究提供了理论依据。
Abstract:
ObjectiveTo predict and explore the key targets and the mechanism of national master of TCM Zhang Lei’s empirical formula for turbidity elimination in the treatment of pulmonary cancer using systems pharmacology and bioinformatics methods. MethodsThe effective compounds and targets of the empirical formula for turbidity elimination were screened via TCMSP; the targets related to pulmonary cancer were gained from GeneCards, mapped with drug action targets to obtain intersecting targets; DAVID database was utilized to perform GO and KEGG pathway enrichment analysis; differential expression analysis was performed using the TCGA database, and molecular docking was carried out between differential genes and the active compounds. ResultsAll 207 active compounds and 162 targets were identified; there are 23 701 targets related to pulmonary cancer, and 51 core action targets were obtained after mapping. DAVID database enrichment analysis demonstrates that the empirical formula for turbidity elimination combats lung cancer possibly through regulating the biological processes including apoptosis, cellular proliferation and gene expressions, and the pathways such as TNF signaling pathway, HIF-1 signaling pathway and PI3K-Akt signaling pathway. "Herb-compound-target-pathway" integrated network reveals that quercetin, luteolin, kaempferol, wogonin, and licochalcone A belong to the key active compounds with a wide range of action. Differential analysis results uncovers that patients with high expressions of BCL2, COL1A1 and TNFRSF1A genes show the lowest overall survival rate compared with the ones with low expressions of the genes (P<0.05); and the differences in the expressions of BCL2 and COL1A1 showed statistical meaning in normal tissue and the tissues of lung cancer (P<0.05); the key targets BCL2 and COL1A1 exhibited good binding activity with the key active compounds. ConclusionThe study primarily uncovers the molecular mechanism of treating lung cancer with turbidity elimination method, unearths that BCL2 and COL1A1 may serve as the key targets for lung cancer treatment, and it could provide theoretical basis for further studies.
