摘要:
目的:观察豆腐果苷(helicid)对坐骨神经慢性压迫伤(CCI)后大鼠的镇痛作用及脊髓c-fos表达的影响.方法:32只Wistar雄性大鼠随机分为4组(n=8);分别为Sham组、CCI组、H1组和H2组。4组分别于CCI手术前1天、手术后第9天进行实验,测后肢的机械痛阈值(MWT)。H1组和H2组手术后10天开始给药,测定1、3、5、7、10天后肢MWT,第14天取大鼠脊髓腰膨大部位免疫组化染色检测Fos免疫阳性细胞。结果:豆腐果苷可明显提高CCI模型大鼠的疼痛阈值(P<0.05),抑制由疼痛所引起的脊髓背角c-fos表达增强的效应(P<0.05)。结论:口服豆腐果苷能剂量依赖性提高CCI大鼠疼痛阈值,降低机械痛觉过敏,抑制神经损伤后脊髓背角c-fos的表达。
Abstract:
Objective: To verify the analgesic effects of helicid on chronic constriction injury of the sciatic nerve (CCI) and c-fos expressions of rats. Method: Thirty-two rats were randomly divided into four groups (n=8):sham group, CCI group, H1 group and H2 group. Four groups were experimented on at the 1st day before CCI surgery and the ninth day after the surgery to detect MWT of hind legs. H1 and H2 groups were given with medicine at the tenth day after the surgery to determine MWT of hind legs at the 1st, 3rd, 5th, 7th and 10th day, at the 14th day lumbar and spinal enlargements were took and fos immunoreactive positive cells were detected with immunohistochemical staining method. Result: Helicid can effectively raise pain threshold of CCI model rats (P<0.05), and inhibit c-fos expressions raise of cornu dorsale medullae spinalis induced by pain. Conclusion: Oral helicid can raise pain threshold of CCI rats dose-dependently, decrease mechanical pain allergy and inhibit c-fos expressions of cornu dorsale medullae spinalis after neural injury.
