摘要:
目的:观察藏药镰形棘豆总黄酮苷元对小鼠的急性毒性作用,以评价该药物的安全性。方法: 以最大给药浓度和最大剂量(0.1 g/kg)灌胃给药,连续观察7天,判断半数致死剂量的可能性,并据此结果,将昆明种小鼠60只分为3组:实验Ⅰ组(0.2 g/kg)、实验Ⅱ组(0.3 g/kg) 和空白对照组(0.5%CMC-Na),连续灌胃给药7天,观察小鼠毒副反应情况,测定出最大耐受量。结果: 小鼠口服镰形棘豆总黄酮苷元无法测出LD50,小鼠最大耐受量为300 g总黄酮苷元/kg(体质量),相当于60 kg(体质量)成人临床生药日用量的378倍。结论: 镰形棘豆总黄酮苷元急性毒性较小,具有一定的使用安全性。
Abstract:
Objective: To assess the safety of LianXing JiDou(Oxytropis Falcata Bunge) through surveying acute toxicity test of total flavonoid aglycones from the drug. Method: The mice received intragastric administration in maximum drug concentration and maximum dose for seven consecutive days, the possibility of lethal median dose (LD-50) was judged. Based on the results, sixty mice were divided into three groups: sample test group I (0.2 g/kg), the test group II (0.3 g/kg) and the control group (0.5% CMC-Na), intragastric administration for seven consecutive days, adverse reactions of the mice were observed to determine maximum tolerance dose. Result: All the mice survived and LD50 could not be measured, maximum tolerance dose of the mice was 300g of total flavonoid aglycones per kilogram, equivalent to 378 times of clinical dose of a man weighing 60 kg. Conclusion: Acute toxicity test shows that total flavonoid aglycones from LianXing JiDou is safe to a certain extent.
