摘要:
目的:观察藏药佐木阿汤对高原红细胞增多症(HAPC)模型大鼠心肌细胞线粒体DNA(mtDNA)及DNA 聚合酶γ等指标表达的影响,探究佐木阿汤对心肌线粒体的保护作用。方法:选用雄性Wistar大鼠32只,随机分为平原对照组、高原模型组、阳性药物组、佐木阿汤组,每组各8只。除平原对照组外,余组均饲养于拉萨以建立HAPC模型,饲养1个月后分别进行生理盐水、红景天和佐木阿汤灌胃干预,干预3个月后取材,采用Realtime PCR、ELISA、Western blot方法分别检测大鼠心肌mtDNA、8-OHdG、DNA聚合酶γ含量。结果:mtDNA含量高原对照组与平原对照组相比,显著降低差异有统计学意义(P<0.05);阳性药物组和佐木阿汤组与高原对照组相比,明显升高(P<0.05);但与平原对照组相比差异有统计学意义(P<0.05)。8-OHdG高原对照组与平原对照组相比,显著增高,差异有统计学意义(P<0.05);阳性药物组和佐木阿汤组与高原对照组相比,显著降低,差异有统计学意义(P<0.05);阳性药物组和佐木阿汤组与平原对照组相比,差异无统计学意义(P>0.05)。DNA聚合酶γ含量高原对照组与平原对照组相比,显著降低差异有统计学意义(P<0.05);阳性药物组和佐木阿汤组与高原对照组相比,差异无统计学意义(P>0.05);阳性药物组和佐木阿汤组与平原对照组相比,无明显增加差异无统计学意义(P>0.05)。结论:佐木阿汤干预HAPC模型大鼠后,心肌mtDNA与DNA聚合酶γ均显著增多,损伤产物8-OHdG显著降低,佐木阿汤可促进线粒体DNA损伤的修复,从而使心肌线粒体的结构和功能逐渐恢复正常,改善HAPC模型大鼠心肌的缺血缺氧状态。
Abstract:
Objective: To explore the mechanism of ZuoMuA Tang protecting myocardial mitochondria by observing its influence on the expressions of mitochondria DNA (mtDNA) and DNA polymerase γ in myocardial cells of rat model with high altitude polycythemia(HAPC). Methods: Thirty-two rats were randomized into plain control group, plateau control group, positive medicine group and ZuoMuA Tang group. Except the rats in plain control group, other rats were kept in Lhasa to prepare HAPC models, and intervened by intragastric administration of physiological saline, HongJingTian(Rhodiola rosea L.) and ZuoMuA Tang after feeding in one months, the tissues were taken after intervened in three months, the contents of myocardial mtDNA, 8-OHdG and DNA polymerase γ of the rats were detected respectively by adopting Realtime PCR, ELISA and Western blot method. Results: The contents of mtDNA decreased notably when plateau control group was compared with plain control group(P<0.05); they raised obviously when positive medicine group was compared with ZuoMuA Tang group and plateau control group (P<0.05); but there was the difference when positive medicine group was compared with plain control group (P<0.05). The levels of 8-OHdG rose significantly when plateau control group was compared with plain control group (P<0.05); they lowered when positive medicine group was compared with ZuoMuA Tang group and plateau control group(P<0.05); there was no obvious difference when positive medicine group was compared with ZuoMuA Tang group and plain control group(P<0.05). The contents of DNA polymerase γ decreased evidently when plateau control group was compared with plain control group(P<0.05); there was no obvious difference when positive medicine group and ZuoMuA Tang group were compared with plateau control group(P>0.05); it rose indistinctly when positive medicine group and ZuoMuA Tang group were compared with plain control group(P>0.05). Conclusion: The levels of myocardial mtDNA and DNA polymerase γ improved significantly, and the levels of damage product 8-OHdG decreased obviously after intervened HAPC rat model by ZuoMuA Tang. ZuoMuA Tang could promote the recovery of mtDNA damage, therefore to make the structure and the functions of myocardial mitochondria restore to normal, and improve ischemic and hypoxia state of HAPC rat model.
