摘要:
目的：对回生胶囊进行小鼠急性毒性实验，并观察回生胶囊与化疗或放疗合用时对S180荷瘤小鼠的抗肿瘤增效、减毒作用，以及对S180荷瘤小鼠免疫功能的影响。方法：急性毒性实验因未测出小鼠致死剂量，无法测定LD50，故改测最大给药量；药效学实验采用回生胶囊8.1、2.7 g生药/kg（体质量）给荷瘤小鼠灌胃。结果：小鼠灌胃给药回生胶囊未发现明显毒性反应，1日最大给药量为42 g/(kg·d)，是临床人日口服剂量0.09 g/(kg·d)的467倍；对环磷酰胺(CTX)化疗或X射线放疗抗小鼠S180肉瘤有增效作用；对CTX或放疗减少S180荷瘤小鼠白细胞和红细胞数有抑制作用；能提高小鼠吞噬细胞的吞噬功能，增强机体非特异性免疫功能；促进小鼠特异性抗体的产生和分泌，增强小鼠的特异性体液免疫功能；促进小鼠脾脏淋巴细胞增殖反应，增强小鼠的特异性细胞免疫功能。结论：回生胶囊1日内过大剂量口服是安全的，与化疗或放疗药物联合应用有增效和减毒作用，同时能增强机体的免疫功能。
Abstract:
Objective: To perform acute toxicity test of HuiSheng capsules on the mice, and to observe synergized and attenuated actions of HuiSheng capsules combined with chemotherapy or radiotherapy on S180 tumor-bearing mice, and its influence on the immunity of S180 tumor-bearing mice. Methods: Lethal doses of the mice didn′t been tested out in acute toxicity test, and LD50 couldn′t been detected, therefore maximum administration dosage has been tested; tumor-bearing mice accepted intragastric administration of HuiSheng capsules in the dosage of 8.1 and 2.7g/kg (body mass) in pharmacodynamic experiment. Results: No obvious toxic reaction has been found when the mice accepted intragastric administration of HuiSheng capsules, maximum administration dosage within one day was 42 g/( kg·d), 467 times of human daily oral dose 0.09 g/( kg·d) in clinic; HuiSheng capsules showed synergized action on S180 tumor of the mice undergoing cytoxan(CTX) chemotherapy or X-ray radiotherapy; it inhibited CTX or radiotherapy induced reduction of white blood cell counting and red blood cell counting of S180 tumor-bearing mice; it raised phagocytic function of phagocytes, enhanced the body′s nonspecific immunity; promoted the production and secretion of specific antibodies, and enhanced specific humoral immune function of the mice; promoted the proliferation of splenic lymphocytes, and enhanced specific cellular immunity of the mice. Conclusion: It′s safe to take HuiSheng capsules in large dose within one day, it shows synergized and attenuated actions when it is combined with chemotherapy or the drugs of radiotherapy, at the same time it could enhance the body′s immunity.
