目的:通过响应面法优化复合酶提取半夏多糖的工艺,并评价其抗氧化活性。方法:以半夏多糖得率为响应值,以液料比、酶解温度、酶解时间、复合酶(木瓜蛋白酶、纤维素酶、果胶酶的质量比为2∶2∶1)添加量为试验因素,采用响应面法建立数学模型,优化提取条件;体外抗氧化活性考察半夏多糖对DPPH和O2-·自由基的清除能力。结果:通过二次回归模型响应面分析,酶解温度、时间、复合酶添加量、液料比4因素对半夏多糖得率的影响依次减弱;酶解温度与时间优化为54℃和57分钟,复合酶添加量为7.2 mg/mL,液料比例为27∶1 mL/g,在此最佳工艺条件下半夏多糖得率为27.98%,模型方程理论预测值为28.32%,两者相对误差小于5%。半夏多糖具有较强的抗氧化活性,对DPPH和O2-·自由基的半数抑制浓度分别为0.987 mg/mL、1.309 mg/mL,但与维生素C比较,抗氧化活性较弱。结论:采用响应面法优化得到了半夏多糖的最佳复合酶提取条件,且半夏多糖有一定的体外抗氧化作用。
Abstract:
Objective: To optimize complex enzymatic extraction technology of polysaccharide from BanXia (Pinellia ternata) by response surface method, and evaluate its antioxidant activity. Methods: Polysaccharide yield of BanXia was taken as the response, liquid to material ratio, enzymatic hydrolysis temperature, enzymatic hydrolysis time and complex enzymes (mass ratios of papain, cellulose and pectase were 2:2:1) as experimental factors, mathematical model was established by adopting response surface method to optimize extraction conditions; antioxidant activity in vitro was used to investigate the clearance ability of polysaccharide extracted from BanXia to DPPH and O2-·radical. Results: By analyzing with two regression model response surface, the influences of enzymatic hydrolysis temperature, enzymatic hydrolysis time, additive amount of complex enzymes and liquid to material ratio became attenuated gradually; enzymatic hydrolysis temperature and enzymatic hydrolysis time were optimized as 54℃ and 57 minutes, additive amount of complex enzymes were 7.2 mg/mL, liquid to material ratio was 27:1 mL/g, under the best extraction technology, polysaccharide yield of BanXia was 27.98%, model equation theoretical prediction value was 28.32%, and relative error between both was less than 5%. Polysaccharide from BanXia presented stronger antioxidant activity, half maximal inhibitory concentrations of DPPH and O2-radical were 0.987 mg/mL and 1.309 mg/mL, but its antioxidant activity was weaker compared with vitamin C. Conclusion: The best complex enzymatic extraction technology of polysaccharide from BanXia is obtained by adopting response surface method, and the polysaccharide from BanXia shows antioxidant activity in vitro to a certain degree.
