摘要:
目的:探讨裸花紫珠总皂苷的最佳提取方案。方法:测定裸花紫珠中总皂苷的含量,并对样品进行测定,通过对不同提取方法和影响因素的比较,得出最佳方案。将样品进行处理后,用5%香草醛冰醋酸溶液、高氯酸、冰醋酸显色,再采用可见分光光度法(检测波长为551 nm)测出吸光度,以三七皂苷为标准品绘制的标准曲线求出总皂苷含量。结果:方法学考察中精密度、重现性、回收率良好,该方法在10~50分钟内稳定,线性范围0~428.4 mg,探究影响因素确定乙醇浓度>乙醇倍量>提取时间。结论:总皂苷的提取工艺中超声工艺优于回流工艺,根据回归曲线测定正交试验中皂苷含量,得出最佳提取方案:用10倍量的75%乙醇常温下超声提取30分钟。
Abstract:
Objective: To study the best extracting method of total saponins from callicarpa nudiflora. Methods: The content of total saponin from callicarpa nudiflora was determined. And the sample was tested. By comparing the different extracting methods and influencing factors, the best extracting method was obtained. After treating the sample, color-reaction was performed with 5% vanillin glacial acetic acid solution, perchloric acid and glacial acetic acid. Then visible spectrophotometry (detection wavelength was 551nm) was adopted to measure absorbance. The content of total saponin was calculated by the standard curve which takes notoginsenoside as the standard sample. Results: Precision, reproducibility and recovery in the study methodology were good, and this method was stable within 10~50 min. ethanol concentration>ethanol volume>extracting time were determined by the study influencing factors. Conclusion: Ultrasonic extraction was better than reflux extraction among the extracting methods of total saponins. Saponin content in orthogonal test was measured according to regression curve for obtaining the best extracting methods. Ultrasonic extraction was performed for 30min at room temperature with 10 times volume 75% ethanol.
