摘要:
目的:探讨胡桃醌和白花丹醌对人结肠癌Caco-2细胞增殖的影响。方法:应用MTT法检测不同浓度和不同作用时间两种萘醌类化合物对Caco-2细胞增殖的抑制作用。结果:胡桃醌在1~100 mol/L浓度范围内,作用48小时,对Caco-2细胞增殖无明显抑制作用,200 mol/L时有显著抑制作用,抑制率为23.3%。白花丹醌在10~100 mol/L浓度范围内分别作用于Caco-2细胞12、24、48、72小时,均可显著抑制细胞增殖,对细胞增殖的半数抑制浓度分别为135、19.89、10.49和4.76 mol/L。结论:胡桃醌对Caco-2细胞增殖的抑制作用较弱,白花丹醌对Caco-2细胞增殖具有较强的抑制作用,且具有时间和剂量依赖性。
Abstract:
Objective: To discuss the effects of juglone and plumbagin on the proliferation of Caco-2 cells of human colon cancer. Methods: The inhibitory actions of juglone and plumbagin in different concentrations and different action times on the proliferation of Caco-2 cells were detected by MTT method. Results: When juglone was ranged from 1 to 100 mol/L, it showed no obvious inhibitory action on the proliferation of Caco-2 cells after acting for 48 hours, it inhibited the proliferation significantly when juglone was in the concentrations of 200 mol/L, and the inhibition rate was 23.3%. When plumbagin was ranged from 10 to 100 mol/L, it showed no obvious inhibitory action on the proliferation of Caco-2 cells after acting for 12 hours, 24 hours, 48 hours and 72 hours respectively, it could notably inhibit cellular proliferation, IC50 for cellular proliferation was 135, 19.89, 10.49 and 4.76 mol/L respectively. Conclusion: Juglone shows poorer inhibitory action on the proliferation of Caco-2 cells, while plumbagin possesses stronger inhibitory action on the proliferation of Caco-2 cells, and the inhibitory action is in a dose and time dependent manner.
