摘要:
目的:观察运脾颗粒对模型大鼠胃肠激素的影响,探讨其防治功能性消化不良(FD)的可能作用机理。方法:采用随机分组法将70只SD大鼠分为正常对照组(空白组)15只,造模组55只,按适度夹尾刺激为主的复合因素建立肝郁脾虚型FD大鼠模型,造模结束后2组各随机抽取5只,并测定胃排空率及小肠推进率,验证造模是否成功。造模结束后将造模组大鼠随机分为模型组、运脾颗粒低、中、高剂量组及西药组,每组10只。空白组及模型组的大鼠每日用生理盐水以0.01 mL/ g灌胃,运脾颗粒高、中、低各剂量组分别配置成0.216 g/mL、0.108 g/mL、0.054 g/mL均以0.01 mL/g灌胃,西药组将多潘立酮片研碎成粉末制成0.27 mg/mL的混悬液以1 mL/100 g灌胃,6组均2次/d,给药14天。干预结束后观察各组大鼠体质量、食量、毛色、激惹情况、精神状态及粪便性状等一般情况;记录实验过程中各组大鼠的体质量、食量,用ELISA法检测各组大鼠血清胃动素(MTL)、胃泌素(GAS)、胆囊收缩素(CCK)含量的变化。结果:与模型组相比,运脾颗粒各剂量组及西药组大鼠进食量、体质量均增加,差异有统计学意义(P<0.01);而运脾颗粒各剂量组与西药组相比,差异无统计学意义(P>0.05)。与模型组相比,运脾颗粒各剂量组及西药组大鼠血清MTL、GAS含量升高,血清CCK含量下降,差异均有统计学意义(P<0.05,P<0.01);且运脾颗粒中、高剂量组大鼠血清MTL、GAS、CCK含量变化与西药组相比,差异具有统计学意义(P<0.05),而低剂量组与西药组相比,差异无统计学意义(P>0.05)。结论:运脾颗粒可增加肝郁脾虚型FD大鼠进食量及体质量,改善模型大鼠的精神状态、粪便、毛色等宏观表征,可能与运脾颗粒上调模型大鼠血清MTL、GAS及下调血清CCK的水平有关,也是运脾颗粒防治FD的作用机理之一。
Abstract:
Objective: To exploring the possible mechanism of spleen-activating granules in the prevention and treatment for functional dyspepsia (FD) by observing its effects on gastrointestinal hormone of the rat model. Methods: Seventy SD rats were allocated to normal control group (the blank group) and 55 rats in model-setting group, FD rat model of liver-depression spleen-deficiency pattern was established by composite factor of mainly clipping tails moderately, after successfully establishing the models, five rats were randomly chosen from both groups respectively, gastric emptying rate and small intestinal propulsive rate were detected to confirm whether the models were successfully established. By the end of modeling, the rats of model-setting group were divided into the model group, low, moderate and high dose groups of spleen-activating granules and domperidone group, ten rats each group. The rats in the blank group and the model group were lavaged with physiological saline each day, 0.01 mL/g, low, moderate and high dose groups of spleen-activating granules received intragastric administration of spleen-activating granules, 0.216 g/mL, 0.108 g/mL and 0.054 g/mL, 0.01 mL/g, domperidone group accepted the suspension, 0.27 mg/mL, 1 mL/100 g after grinding domperidone, six groups were medicated for 14 days, twice each day. General conditions including fecal character, mental state, irritation, coat colour, appetite and body weight of the rats in different groups were observed by the end of the intervention; the contents of MTL, GAS and CCK in the serum of the rats in different groups were measured by ELISA method after recording body weight and appetite of the rats during the experimental course. Results: Compared with the model group, the appetite and body weight of the rats in different dose groups of spleen-activating granules and western medicine group increased, and the difference had statistical meaning (P<0.01); while the difference had no statistical meaning when different dose groups of spleen-activating granules were compared with western medicine group (P>0.05). Compared with the model group, the contents of MTL and GAS raised in different dose groups of spleen-activating granules and western medicine group, the contents of CCK decreased, and the difference had statistical meaning (P<0.05, P<0.01); and the difference was statistically significant in the comparisons of the contents of MTL, GAS and CCK when moderate and high dose groups of spleen-activating granules were compared with western medicine group (P<0.05), and the difference had no statistical meaning when low dose group was compared with western medicine group (P>0.05). Conclusion: Spleen-activating granules could increase food intake and body weight of the rats of FD of liver-depression spleen-deficiency pattern, improve mental state, feces, coat colour of the models, which might be related to that it could up regulate the content of MTL and GAS of the model and down regulate the level of CCK in the serum, and it′s one of the mechanisms of spleen-activating granules in the prevention and treatment for FD.
