摘要:
目的:观察木犀草素对酒精所致小鼠急性肝损伤的保护作用。方法:将120只雄性小鼠随机分成空白对照组、模型对照组、阳性对照组[联苯双酯150 mg/(kg·d)、木犀草素低剂量组(25 mg/kg)、木犀草素中剂量组(50 mg/kg)、木犀草素高剂量组(100 mg/kg)],采用酒精灌胃方法分别建立小鼠醉酒和急性肝损伤模型,观察木犀草素对小鼠生理状态、肝指数及血清中丙氨酸氨基转移酶(ALT/GPT)、天冬氨酸氨基转移酶(AST/GOT)、甘油三酯(TG)和胆固醇(TC)的影响,同时通过苏木精-伊红染色(HE染色)光学显微镜观察小鼠肝组织病理学改变及试剂盒检测超氧化物歧化酶(SOD)、丙二醛(MDA)变化,考察木犀草素对急性酒精性肝损伤的保护的作用及可能机制。结果:与模型组比较,木犀草素可以改善小鼠醉酒状态,降低小鼠肝指数,给药组血清学指标ALT/GPT、AST/GOT、TG、TC降低,病理组织学检查结果表明木犀草素对小鼠肝小叶结构、肝细胞坏死和变性及炎细胞浸润等方面均有改善作用,且肝组织内SOD升高,MDA降低。结论:木犀草素具有防醉和缓解酒精所致的肝损伤的作用,其机制可能与降低自由基生成、减轻脂质过氧化有关。
Abstract:
Objective: To observe the protective effects of luteolin against acute liver injury(ALI) induced by alcohol of the mice. Methods: All 120 mice were randomly allocated to blank control group, model control group, positive control groups [biphenyl dicarboxylate 150 mg/( kg·d) , low dosage group of leteolin (25 mg/kg), moderate dosage group of luteolin(50 mg/kg) and high dosage group of leteolin (100 mg/kg)]. Drunk mice model and ALI mice model were established respectively by using lavage of alcohol, to observe the effects of luteolin on physiological state, liver index, ALT/GPT, AST/GOT, TG and TC, simultaneously, the changes of SOD and MDA were detected by kit, pathological changes of liver tissue in the mice were observed by HE staining method under light microscope, the protective effects of luteolin against ALI induced by alcohol and the potential mechanism were investigated.Results: Compared with the model group, luteolin could improve the drunk state of the mice, decrease liver index of the mice, serological indexes in the medication groups including ALT/GPT, AST/GOT, TG and TC decreased, the results of histopathologic examination demonatrated that luteolin could improve hepatic lobule structure, necrosis, degeneration of liver cells, inflammatory cell infiItration of the mice, SOD raised and MDA lowered in liver tissue. Conclusion: Luteolin possesses the effects of preventing drunkness and relieving alcohol-induced ALI, and its mechanism might be related to decreasing free radical formation and relieving lipid peroxidation.
