摘要:
目的:建立一种稳定、可靠的大鼠局灶性脑缺血再灌注模型制备方法,并评价其效果。方法:用线栓阻塞SD大鼠(260~280 g)右侧大脑中动脉,2小时后再灌注,术后2小时及第3、7、14天进行神经功能评分,并通过脑组织2,3,5-氯化三苯基四氮唑(TTC)染色对模型进行评价。结果:模型组大鼠死亡率为29.6%,术后2小时,3、7、14天神经行为学评分分别为(1.94±0.80)分、(1.61±0.78)分、(1.50±0.67)分及(1.33±0.52)分,均显著高于假手术组(P<0.01)。经TTC染色发现病灶范围较恒定,术后3、7、14天脑梗死面积占整个脑面积百分比分别为(32.61±0.76)%,(29.50±4.03)%和(13.32±3.24)%,与假手术组同期比较均有显著性差异(P<0.01)。结论:采用西浓线栓制备过程中注重细节完善,便可得到稳定的SD大鼠局灶性脑缺血再灌注模型。
Abstract:
Objective: To introduce a stable and reliable preparation method for focal cerebral ischemia reperfusion rat model, and assess the effects. Methods: Right middle cerebral artery of SD rat was occluded, and reperfused in two hours, neurological function score was performed in two hours, three days, seven days and 14 days after the operation, and the model was assessed by TTC staining in cerebral tissue. Results: Mortality rate of the rats in the model group was 29.6%, neurological behavior scores of them in two hours after the operation, three days, seven days and 14 days were(1.94±0.80) points, (1.61±0.78) points, (1.50±0.67) points and (1.33±0.52) points, remarkably higher than these of sham operation group (P<0.01). After TTC staining, the range of foci was constant, the percentages of cerebral infarction area occupying the entire brain area were (32.61±0.76)%, (29.50±4.03)% and (13.32±3.24)%, there was the significance difference when they were compared with these of sham operation group (P<0.01). Conclusion: Stable rat model with focal cerebral ischemia reperfusion could be obtained by applying intraluminal thread to SD rats and paying attention to perfect details in the preparation courses.
