摘要:
目的:探讨紫草素(Shikonin)对荷肝癌大鼠免疫功能的影响及其作用机制。方法:采用腋窝皮下注射肝癌Walker-256细胞的方法建立荷肝癌移植瘤大鼠模型,设紫草素[40、20、10 mg/(kg·d)]组和顺铂[2 mg/(kg·d)]组(n=20),腹腔注射给药,疗程14天。观察大鼠瘤体生长状况并计算抑瘤率,苏木精-尹红(HE)染色观察瘤体组织形态结构;噻唑蓝(MTT)比色法测定脾淋巴细胞转化率;酶联免疫吸附法(ELISA)测定血清白细胞介素2(IL-2)和肿瘤坏死因子α(TNF-α);流式细胞术(flow cytometry,FCM)检测血液T淋巴细胞亚群CD4+、CD8+百分率并计算CD4+/CD8+比值。结果:与模型组比较,紫草素组荷肝癌大鼠瘤体缩小且硬度降低,瘤体组织呈现细胞皱缩、组织片状坏死等病理性改变;紫草素各剂量组抑瘤率升高(P<0.05);紫草素[40、20 mg/(kg·d)]组脾淋巴细胞转化率升高(P<0.05),血清中IL-2、TNF-α含量降低(P<0.05),紫草素[40 mg/(kg·d)]组大鼠血液中CD4+百分率升高(P<0.05)、紫草素[40、20 mg/(kg·d)]组大鼠CD8+降低且CD4+/CD8+比值升高(P<0.05)。结论:紫草素具有提高荷肝癌大鼠免疫功能的作用;其机制可能与紫草素能够有效提高脾淋巴细胞转化率,降低IL-2、TNF-α含量以及提高CD4+百分率、降低CD8+百分率而提高CD4+/CD8+比值有关。
Abstract:
Objective: To explore the influence of Shikonin on immunologic function of the rats suffering liver cancer and its mechanism. Methods: Rat model suffering transplanted tumor of liver cancer were established by hypodermic injection of liver cancer Walker-256 at the armpit, the groups of Shikonin[40, 20 and 10 mg/( kg·d)] and cis-platinum complexes group [2 mg/( kg·d)](n=20) were established by peritoneal injection, and the course was 14 days. Growth conditions of the tumor of the rats were observed, and tumor-inhibitory rate was calculated, histological and morphological structures of the tumor were observed by HE staining; splenic lymphocyte transformation rate was measured by MTT chromometry; the levels of IL-2 and TNF-α were detected by ELISA method; the percentages of CD4+ and CD8+ T lymphocyte subsets by flow cytometry(FCM), and the ratio of CD4+/CD8+ was calculated. Results: Compared with the model group, tumor body shrank and the hardness lowered of the rats after intervened by Shikonin, tumor tissue showed pathological changes including cell shrinking, flake necrosis of the tissue; tumor-inhibitory rate in different intervention groups of Shikonin lifted(P<0.05); splenic lymphocyte transformation rates in the groups of Shikonin [40, 20 and 10mg/( kg·d)] raised (P<0.05); the contents of IL-2 and TNF-α decreased (P<0.05), the percentage of CD4+ in the blood of Shikonin group [40 mg/(kg·d)] evaluated (P<0.05), CD8+ in Shikonin group[40 and 20 mg/(kg·d)] lowered while the ratio of CD4+/CD8+ raised (P<0.05). Conclusion: Shikonin could improve immunologic function of the rats suffering liver cancer, and its mechanism might be related to that Shikonin could effectively raise splenic lymphocyte transformation rate, lower the contents of IL-2 and TNF-α, evaluate CD4+ percentage, decrease CD8+ percentage while lift the ratio of CD4+/CD8+.
