摘要:
目的:探索肠瘤康基于p16/CyclinD1/CDK4/Rb通路对人结肠癌HT29细胞周期的作用及机制。方法:取对数生长期的人结肠癌HT29细胞,待细胞完全贴壁后加入梯度浓度的肠炎原溶液,分为对照组(肠瘤康0 μg/mL)、肠瘤康高(100 μg/mL)、中(10 μg/mL)、低(0.0 μg/mL)剂量组及5-氟尿嘧啶组(5氟尿嘧啶4 μg/mL)通过MTT法检测肠瘤康对HT29细胞的增殖抑制作用,流式细胞仪检测肠瘤康对HT29细胞周期的影响,实时定量PCR法检测细胞内p16、CyclinD1、CDK4 及Rb基因表达水平。结果:肠瘤康干预后,HT29细胞的增殖抑制率显著性降低,流式细胞术检测发现G1期的百分比显著增加。肠瘤康可以剂量依赖性的抑制结肠癌HT29细胞增殖,促进p16 mRNA表达量,抑制CyclinD1、CDK4、Rb mRNA的表达量。结论:肠瘤康能抑制HT29细胞的增殖,诱导细胞在G1期阻滞,其诱导作用与其调控p16/CyclinD1/CDK4/Rb通路上相关因子表达有关。
Abstract:
Objective:To explore the effects of ChangLiuKang on the cycle of human colon cancer HT29 cells based on p16/CyclinD1/CDK4/Rb pathway and the related mechanism. Methods: Human colon cancer HT29 cells were chosen at logarithmic growth phase, after the adherent cells were obtained, the solution of enteritis proto in gradient concentrations were added, the cells were separated into the control group(Changliakag 0 μg·mL-1), high (100 μg·mL-1), moderatel(10 μg·mL-1) and 10W(0.0 μg·mL-1) dosed groups of ChangLinKang and 5-fluorouracil group(5-fluorouracil 4 μg·mL-1). The inhibitory effects of ChangLiukang on the proliferation of HT29 cells were detected by MTT method, the influence of ChangLiuKang on the cycle of HT29 cells were measured by flow cytometry, and the expressions of p16, CyclinD1, CDK4 and Rb genes in the cells by real-time quantitative(RTQ) PCR method. Results: After intervened by ChangLiuKang, proliferation inhibitory rate of HT29 cells decreased notably, the results of flow cytometry showed the percentage of G1 stage increased remarkably. ChangLiuKang could inhibit the proliferation of colon cancer HT29 in a dose-dependent manner, promote the expression of p16 mRNA, inhibit the expression of CyclinD1, CDK4 and Rb mRNA. Conclusion:ChangLiuKang could inhibit the proliferation of HT29, and induce the cells arresting at G1 stage, and its inducing effects are related to regulating the expressions of the associated factors of p16/CyclinD1/CDK4/Rb pathway.
