摘要:
目的:探讨洋葱皮提取物对离体蟾蜍心肌收缩力和心率的影响及其机制。方法:根据斯氏蛙心插管方法,分别用浓度为0.02、0.04、0.06、0.08、0.10 mg/mL的洋葱皮提取物灌流离体蟾蜍心脏,采用BL-420生物机能实验系统记录心率和心肌收缩力的变化情况;分别加入低钙、正常钙、高钙任氏液和0.000 5 mg/mL维拉帕米,5分钟后充分灌洗,再分别加入低钙、正常钙、高钙任氏液配制的洋葱皮提取物以及0.000 5 mg/mL维拉帕米与洋葱皮提取物的混合物,记录5分钟心率和心肌收缩力的变化情况。结果:灌流0.02、0.04、0.06、0.08、0.10 mg/mL的洋葱皮提取物的离体蟾蜍心率变化不大(P>0.05),但离体蟾蜍心肌张力峰峰值先逐渐升高,约1分钟达高峰,而后逐渐下降,5分钟时仍高于加药前心肌张力峰峰值(P<0.05),表明蟾蜍心收缩力呈现出典型的先升高后下降的趋势,且具有明显的药物浓度依赖性;0.000 5 mg/mL维拉帕米、低钙任氏液对蟾蜍心脏具有负性肌力的作用,且洋葱皮提取物能够拮抗其对心肌的抑制作用,高钙任氏液对离体心脏具有正性肌力作用,而洋葱皮提取物能够增强其对心脏的正性肌力作用。结论:洋葱皮提取物可使离体蟾蜍心脏心肌收缩力先升高后下降,且有药物浓度依赖性,但是对心率影响不大,且其兴奋心肌的作用机制可能与激活L-型钙通道有关。
Abstract:
Objective: To explore the effects of onion skin extract on heart rate and myocardial contraction force of isolated toad heart and its mechanism. Methods: According to steinmann's frog heart intubation, 0.02, 0.04, 0.06, 0.08 and 0.10mg/mL onion skin extract were used to perfuse isolated frog heart respectively, the changes of heart rate and myocardial contraction force were recorded using BL-420 biological function experimental system; after adding low, normal and high calcium ringer solution and 0.000 5mg/mL verapamil respectively, the hearts were perfused completely in five minutes, and then adding onion skin extract prepared by low, normal and high calcium ringer solution and the mixture including 0.000 5 mg/mL verapamil and onion skin extract, the changes of heartrate and myocardial contraction force were recorded within five minutes. Results: 0.02, 0.04, 0.06, 0.08 and 0.10 mg/mL onion skin extract couldn't change heart rate of isolated toad heart significantly (P>0.05), while the value of myocardial stretch peak rose gradually, reached the summit around one minute, decreased gradually, the value in five minutes were still higher than the peak value before medication (P<0.05), contraction force of toad heart presented the typical tendency of first rising and then decreasing in the dose-dependent manner obviously; 0.000 5 mg/mL verapamil and ringer solution containing low dose of calcium owned negative muscle force, and onion skin extract could fight against their inhibitory effects on cardiac muscle, high calcium ringer solution possessed positive muscle force on isolated heart, while onion skin extract could enhance the positive effects on heart. Conclusion: Onion skin extract could first raise and then lower myocardial contraction force of isolated toad heart in dose-dependent manner, but show little effects on heart rate, and the mechanism of cardiac muscle stimulation might be related to the activation of L-type calcium channels.
