摘要:
目的：探讨藏药二十五味儿茶凝胶对慢性湿疹模型大鼠及其血清白细胞介素4(IL-4)、白细胞介素10(IL-10)、干扰素γ(IFN-γ)的影响。方法：将72只SD大鼠随机分为2组，空白组12只，另一组共60只造模成功后随机分为模型对照组、冰黄肤乐组及藏药二十五味儿茶凝胶高、中、低剂量组。冰黄肤乐组给予冰黄肤乐，藏药二十五味儿茶凝胶低(0.14 g/kg)、中(0.21 g/kg)、高(0.35 g/kg)剂量组经皮给药，给药4周后观察皮肤外观、病理变化及症状积分，并采用 ELISA法检测大鼠血清中IL-4、IL-10、IFN-γ的含量。结果：藏药二十五味儿茶凝胶可改善慢性湿疹大鼠角化表皮、增厚棘细胞层、延长表皮突等，各给药组与模型对照组相比差异有统计学意义(P＜0.05)；各给药组与模型对照组苔藓化积分比较，差异有统计学意义(P＜0.05)，红斑与丘疹积分比较，差异无统计学意义(P＞0.05)；与空白组比较，模型对照组大鼠血清IL-4、IL-10含量升高，IFN-γ含量降低，差异有统计学意义(P＜0.05)；与模型对照组比较，藏药二十五味儿茶凝胶低、中、高剂量组血清IL-4、IL-10含量降低，IFN-γ含量升高，差异有统计学意义(P＜0.05)，低剂量组与冰黄肤乐组比较差异无统计学意义(P＞0.05)。结论：藏药二十五味儿茶凝胶能有效改善慢性湿疹红斑、丘疹、苔藓化症状，其作用机制可能与免疫调节及减少炎症反应有关。
Abstract:
Objective: To explore the effects of Tibetan medicine ErShiWuWei catechu gel on IL-4, IL-10 and IFN-γ of rat model with chronic eczema. Methods: All 72 SD rats were randomized into two groups, 12 rats in the blank group, 60 rats in another group were randomized into model control group, BingHuang FuLe group, high, moderate and low doses groups of Tibetan medicine ErShiWuWei catechu gel after they were prepared into the models suffering chronic eczema successfully. BingHuang FuLe group were given BingHuang FuLe, high, moderate and low doses groups accepted the gel through transdermal administration in different concentrations: 0.35, 0.21 and 0.14 g/kg, after medicating for four weeks, symptoms scores, pathological changes and skin appearance were observed, the contents of IL-4, IL-10 and IFN-γ of rats were detected using ELISA method. Results: Tibetan medicine ErShiWuWei catechu gel could improve pathological manifestations including epidermal keratosis, spinous cell thickening, extension of epidermal process and others, the difference was statistically significant when different medication groups were compared with model control group (P＜0.05); the difference showed statistical meaning when the treatment group was compared with model control group in lichenification integrals (P＜0.05), the difference had no statistical meaning in the comparison of erythema and papula scores (P＞0.05); compared with the blank group, the contents of IL-4 and IL-10 in model control group rose while the contents of IFN-γ lowered, and the difference showed statistical meaning (P＜0.05); compared with model control group, the contents of IL-4 and IL-10 decreased while the contents of IFN-γ increased in low, moderate and high doses groups of ErShiWuWei catechu gel, and the difference showed statistical meaning (P＜0.05), no difference had been found in the comparisons of low dose group and BingHuang FuLe group (P＞0.05). Conclusion: Tibetan medicine ErShiWuWei catechu gel could effectively improve erythema, papula and lichenification of chronic eczema, and its mechanism might be related to immunoregulation and reducing inflammatory reactions.
