摘要:
目的:观察四君子汤合膈下逐瘀汤对人肝癌细胞HepG2和SMMC-7721的影响。方法:倒置显微镜观察中药组、阴性对照组、氟尿嘧啶组中HePG2及SMMC-7721细胞生长情况。采用MTT法检测3组细胞生长的抑制率,流式细胞仪(FCM)检测人肝癌HepG2和SMMC-7721细胞凋亡情况及细胞周期。结果:倒置显微镜下观察阳性对照组及中药组中人肝癌HepG2和SMMC-7721细胞生长明显受抑制,MTT法测定细胞增殖抑制率。中药对HepG2和SMMC-7721细胞的增殖均有明显的抑制作用,与阴性对照组比较差异有统计学意义(P<0.05)。FCM检测中药对HepG2、SMMC-7721细胞凋亡具有一定的诱导作用,与对照组比较差异有统计学意义(P<0.05)。结论:四君子汤合膈下逐瘀汤具有抑制人肝癌HepG2和SMMC-7721细胞增殖,促进其凋亡的作用。
Abstract:
Objective:To observe the effects of SiJunZi decoction combined with GeXia ZhuYu decoction on human liver cell line HepG2 and SMMC-7721. Methods: Inverted microscope was used to observe the growth conditions of SMMC-7721 and HePG2 in herbal group, negative control group and fluorouracil group. MTT method was adopted to detect the inhibitory rate of cellular growth in three groups, flow cytometry (FCM) was used to detect the apoptosis conditions of human liver cell HepG2 and SMMC-7721 and cell cycles. Results: The growth of human liver cell line HepG2 and SMMC-7721 cells were obviously inhibited in the positive control group and herbal group under the inverted microscope, MTT method was used to detect the inhibitory rate of cellular proliferation, herbs showed the notable inhibitory effects on the proliferation of HepG2 and SMMC-7721 cells, there was the significant difference when compared with negative control group(P<0.05). FCM detection demonstrated that herbs could induce cellular apoptosis of HepG2 and SMMC-7721 to a certain degree, there was the significant difference when compared with the control group (P<0.05). Conclusion: SiJunZi decoction combined with GeXia ZhuYu decoction could inhibit the proliferation of human liver cell line HepG2 and SMMC-7721 and induce their apoptosis.
