摘要:
目的:探讨敦煌遗方“神仙粥”对阿尔兹海默病(AD)模型大鼠学习记忆能力及海马组织Bcl-2、Bax基因表达的影响。方法:购置48只SPF级健康SD大鼠,雌雄各半,2月龄,体质量(150±30)g,从中随机抽取出8只作为空白组,其余40只大鼠皮下注射D-半乳糖制作AD模型[125 mg/(kg·d)],空白组则注射与之等剂量的生理盐水。造模成功后,将40只AD模型大鼠随机分为模型组,阳性对照组,神仙粥低、中、高剂量组,每组8只,雌雄各半,进行干预实验。采用Morris水迷宫实验检测大鼠学习记忆能力,免疫组织化学检测海马组织Bcl-2、Bax基因表达情况。结果:1)各组大鼠定向航行试验平均逃避潜伏期在不同时点与不同组别维度差异均有统计学意义(P<0.05),且后续两天较之于第一天组大鼠定向航行试验平均逃避潜伏期缩短;此外,神仙粥高、中、低组与模型组和空白组差异均有统计学意义(P<<0.05),且平均逃避潜伏期均高于空白组,低于模型组。2)各组在目的象限停留时间、有效区域停留时间、平台停留时间上差异均有统计学意义(P<0.05),且神仙粥高、中、低组在目的象限停留时间和有效区域停留时间均大于模型组,神仙粥高、中组以及空白组在平台停留时间均大于模型组。3)各组大鼠海马组织Bcl-2比较,空白组与其余各组差异均有统计学意义(P<0.05),且值均低于其余各组;模型组与阳性对照组、神仙粥高剂量组差异均有统计学意义(P<0.05),且值低于其余2组;阳性对照组与神仙粥中、低剂量组差异均有统计学意义(P<0.05),且值高于后两者;各组大鼠脑皮质Bax比较,空白组与其余各组差异均有统计学意义(P<0.05),且值均低于其余各组;模型组与阳性对照组、神仙粥高剂量组差异均有统计学意义(P<0.05),且值高于其余2组。结论:敦煌遗方神仙粥可通过调节海马组织Bcl-2、Bax基因表达来抑制细胞凋亡,从而改善AD模型大鼠的学习记忆能力,具有一定的抗衰老作用。
Abstract:
Objective:To discuss the effects of DunHuang YiFang "ShenXian Zhou" on learning and memory ability, the expressions of Bcl-2 and Bax gene in the hippocampus of AD rat model. Methods: All 48 SPF SD rats, two months old, weighing (150±30)g, were bought, and eight rats were randomly chosen as the blank group, the other rats accepted subcutaneous injection of D-galactose to prepare AD model [125 mg/(kg·d)], and the rats in the blank group were injected with equivalent dose of physiological saline. After successfully preparing the models, AD rat models were randomized into the model group, positive control group, low, moderate and high dose groups of ShenXian Zhou, eight rats each group, half male and female, who accepted intervention experiments, learning and memory ability of the rats were detected using Morris water maze test, the expressions of Bcl-2 and Bax gene in the hippocampus were measured by immunohistochemical method. Results: 1)The difference had statistical meaning in mean escape latency at different time points between different groups in directional navigation test(P<0.05) mean escape latency shortened in the following two days compared with the first day, desides the differences had statistical meaning when low, mode rate and high closes groups of ShenXian Zhou were, compared with the model group and the blank group(P<0.05). Time point dimensions: Mean escape latency of the rats in different groups at the first day were compared with these at the second and third day, and the difference was statistically significant(P<0.05), mean escape latency of the rats in directional navigation test at the second and third days shortened compared with the first day; the dimension of groups: the difference showed statistical meaning when the model group was compared with the others (P<0.05), and mean escape latency was higher than others of the other group. The comparisons of space exploration time among different groups, when the model group was compared with the others, the difference had statistical meaning in the retention time of purpose quadrant, effective area and platform(P<0.05), and the retention times were shorter than the others. The results of the comparisons of Bcl-2 in the hippocampus among different groups showed: there were a significant difference when the blank group was compared with the other groups(P<0.05), and the expression values were lower than these of the other groups, when the model group was compared with positive control group and high dose group of ShenXian Zhou, the differences had statistical meaning (P<0.05), and the expressions were lower than these of the other two groups, when the positive control group was compared with low and moderate dose groups of ShenXian Zhou, the differences were found in the comparisons (P<0.05), and the expressions were higher than the other two groups; the results of the comparisons of Bax expressions in the pallium between different groups, when the blank group was compared with the others, the difference showed statistical meaning (P<0.05), the values were lower than these of the other groups, the difference had statistical meaning when the model group was compared with positive control group and high dose group of ShenXian Zhou(P<0.05), and the value was higher than these of the other two groups. Conclusion: DunHuang YiFang "ShenXian Zhou" could improve learnning and memory ability of AD rat model, possess certain anti-aging effects by regulating the expressions of Bcl-2 and Bax gene, therefore to inhibit cellular apoptosis.
