摘要:
目的：探讨番茄红素对人非小细胞肺癌细胞A549凋亡及对细胞周期的影响。方法：体外培养并取处于对数生长期的A549细胞进行分组，分别给予培养液（空白对照组）、不同浓度番茄红素（20、10、5 μg/mL）及顺铂（40 μg/mL）进行干预，每组10个复孔。药物干预48小时后，噻唑蓝（MTT）比色法测定各组细胞增值抑制率；流式细胞术（FCM）检测各组细胞周期、观察细胞凋亡状况并计算凋亡率；应用逆转录-聚合酶链反应（RT-PCR）法检测细胞凋亡相关基因Bcl-2 mRNA、Bax mRNA表达；蛋白免疫印迹（Western blotting）法检测细胞周期蛋白（cyclin B1、cyclin D1）和周期蛋白依赖性激酶（CDK1、CDK2）表达。结果：与空白对照组比较，番茄红素各剂量组和顺铂组A549细胞增值抑制率提高，番茄红素（20、10 μg/mL）组细胞处于G2-M期的比例提高、凋亡率显著升高，Bcl-2 mRNA表达下调且Bax mRNA表达上调，Bax/Bcl-2值显著提高，cyclin B1蛋白和CDK1蛋白表达上调，cyclin D1蛋白和CDK2蛋白表达下调，差异均具有统计学意义（P＜0.05）。结论：番茄红素具有促进人非小细胞肺癌A549细胞凋亡，阻滞其有丝分裂，从而抑制其增殖的作用，可能与番茄红素影响凋亡相关调控基因及细胞周期相关调控蛋白表达有关。
Abstract:
Objective: To discuss the effects of lycopene on the cycle and the apoptosis of human non-small cell lung cancer (NSCLC) A549 cells. Methods: After cultured in vitro, A549 cells at logarithmic growth phase were chosen and grouped, they were given nutrient solution (blank control group), different concentrations of lycopene (20, 10 and 5 μg/mL) and cis-platinum complexes (40 μg/mL) to intervene, ten compound holes each group. After drug intervention for 48 hours, MTT chromatometry was used to detect the inhibitory rates of cellular proliferation in different groups; flow cotymetry (FCM) was adopted to measure cellular cycles in different groups, observe apoptosis conditions and calculate the apoptosis rate; RT-PCR method was applied to detect the expressions of cellular apoptosis - related genes Bcl-2 mRNA and Bax mRNA; Western blotting method was used to measure the expressions of cyclin B1, cyclin D1, CDK1 and CDK2. Results: Compared with blank control group, the inhibitory rate of A549 cells proliferation in different doses groups of lycopene and cis-platinum complexes group increased, the ratio of the cells in the groups of lycopene (20 and 10 μg/mL) at G2-M phase increased, apoptotic rate lifted notably, the expressions of Bcl-2 mRNA up regulated, Bax/Bcl-2 value upped remarkably, the expressions of cyclin B1 protein and CDK1 protein up regulated, the expressions of cyclin D1 protein and CDK2 protein down regulated, and the difference showed statistical meaning(P＜0.05). Conclusion: Lycopene could promote the apoptosis of NSCLC A540 cells, block the mitosis and inhibit its proliferation, which might be related to that lycoypene could influence on the expression of apoptosis - related regulatory gene and cellular cycle related regulatory protein.
