摘要:
目的：从抗炎、调节磷酸化ERK1/2蛋白表达方面来探讨黄连素抗幽门螺旋杆菌（Hp）相关性胃炎胃黏膜损伤、保护胃黏膜的作用机制。方法：建立Hp相关性胃炎大鼠模型48只，随机分成正常组、正常+黄连素组、模型组和模型+黄连素组，每组12只。8周后，ELISA法检测胃黏膜组织白介素6(IL-6)、白介素10(IL-10)、肿瘤坏死因子α(TNF-α)的含量。Wester Blot法检测白介素10(IL-10)、白介素1β(IL-1β)、细胞外调节信号蛋白激酶(ERK1/2)的蛋白含量。结果：黄连素能显著降低HP相关性胃炎大鼠胃黏膜的IL-6、IL-1β、TNF-α、磷酸化ERK1/2蛋白含量，增加IL-10的含量。结论：黄连素可通过减少模型大鼠促炎细胞因子(TNF-α、IL-1β、IL-6)，增加抗炎细胞因子(IL-10)的释放，恢复致炎因子(PICs)和抗炎因子(AICs)之间的平衡,抑制Hp引起的炎症反应对胃黏膜的损伤，保护胃黏膜。
Abstract:
Objective: To discuss the mechanism of berberine resisting gastric mucous membrane injury and protecting gastric mucosa of Hp-related gastritis from the aspects of anti-inflammation and regulating ERK1/2 protein expressions. Methods: After establishing 48 Hp-related gastritis rat model, the rat models were randomized into the normal group, normal+berberine group, the model group and the model+berberine group, 12 rats each week. In eight weeks, ELISA method was used to detect the contents of IL-6, IL-10 and TNF-α in gastric mucosa, Western blot method was adopted to measure the contents of IL-10, IL-1β and ERK1/2 protein. Results: Berberine could significantly reduce the contents of IL-6, IL-1β, TNF-α, phosphorylated ERK1/2 protein of gastric mucosa in the rats, and increase the contents of IL-10. Conclusion: Berberine could inhibit gastric mucous membrane injury caused by Hp-induced inflammatory reaction and protect gastric mucosa through reducing TNF-α, IL-1β and IL-6, increasing the release of IL-10 and restore the balance between PICs and AICs.
