摘要:
目的:探究钩吻乙醇提取液对由脂多糖半乳糖胺导致的急性肝衰竭大鼠模型的保护作用。方法:将Wistar雄性大鼠随机分为3组,正常组、模型组、实验组各5、10、11只。实验组给予钩吻乙醇提取物灌胃,0.5小时后腹腔注射D-半乳糖胺(D-GalN),1小时后腹腔注射脂多糖(LPS);模型组直接腹腔注射D-GalN,正常组腹腔注射NS。1小时后各组眼眶取血用于后备测量血清肿瘤坏死因子α(TNF-α),实验组4小时后再给予钩吻乙醇提取物灌胃1次。12小时后各组腹主动脉取血,剖取肝脏病理组织完成实验标本采集,比较各组大鼠血清中丙氨酸氨基转移酶(ALT)、总胆红素(TBIL)、一氧化氮(NO)、TNF-α的差异,检测各组大鼠肝脏组织中丙二醛(MDA)的变化,观察各组大鼠造模完成后一般体征的差异和各组大鼠肝脏HE染色病理切片的不同形态差异。结果:模型组、实验组与正常组比较大鼠的活动度减弱,精神萎靡抗捕捉能力差;血清ALT、TBIL、NO和TNF-α均显著升高(P<0.05);模型组和实验组比较差异无统计学意义(P>0.05)。结论:钩吻乙醇提取液对脂多糖半乳糖胺导致的Wistar大鼠急性肝衰竭未发现明显的保护作用,钩吻具有急性肝脏毒性。
Abstract:
Objective: To survey the protective effects of elegant jessamine ethanol extract on the rats with lipopolysaccharide galactosamine- induced acute hepatic failure. Methods: All Wistar male rats were randomized into three groups: 5 rats in normal group, 10 rats in the model group and 11 rats in the experiment group. The experiment group were drenched with elegant jessamine ethanol extract, and accepted peritoneal injection of D-GalN in half an hour, and LPS in one hour; the model group received peritoneal injection of D-GalN directly, and the normal group peritoneal injection of NS. Orbital blood samples were drawn in one hour to prepare the detection of TNF-α, and the experiment group were drenched with elegant jessamine ethanol extract once in four hours. The blood was taken from aorta abdominalis in 12 hours, and the pathological tissues of liver were cut out to finish the collection of experimental samples, to compare the differences of ALT, TBIL, NO and TNF-α of the serum in the rats of different groups, detect the changes of MDA in liver tissue of the rats in different groups, and observe the differences of general body signs of the rats in different groups after modeling and pathological slices of liver tissues. Results: When the model group and the experiment group were compared with the noraml group, activity of the rats weakened, mental malaise anti-capture ability was poor; the levels of ALT, TBIL, NO and TNF-α raised significantly (P<0.05); the difference between the model group and the experiment group was insignificant (P>0.05). Conclusion: Elegant jessamine ethanol extract shows no obvious protective effects on the rats, and elegant jessamine owns acute liver toxicity.
