摘要:
目的:研究马铃薯提取物糖苷生物碱、马铃薯汁、马铃薯泥对化疗性静脉炎兔模型纤维蛋白溶解酶原激活剂(tissue-type plasminogen activator,t-PA)和1型纤维蛋白溶解酶原激活抑制剂(plasminogen activator inhibitor-1,PAI-1)表达及兔耳注射部位温度的影响。方法:将60只普通级日本大耳白兔随机分为空白对照组、模型组、硫酸镁组、糖苷生物碱组、马铃薯汁组及马铃薯泥组,通过耳缘静脉推注长春瑞滨建立兔静脉炎模型,造模后在各组兔穿刺部位涂抹相应药物,在相应时间点检测兔耳注射部位温度,免疫组化法检测t-PA及PAI-1含量。结果:1)造模后兔耳温度与造模前比较升高(P<0.01),涂药第2天糖苷生物碱组、第3天硫酸镁组、马铃薯泥组、糖苷生物碱组与同组造模后及同时间点模型对照组比较兔耳温度下降,差异有统计学意义(P<0.05)。2)t-PA、PAI-1的表达:空白对照组兔耳组织镜下可见t-PA、PAI-1少量表达,呈棕黄色颗粒;模型组血管周围组织及血管内血栓中呈强阳性表达(P<0.01);与模型组比较,除马铃薯汁组PAI-1表达差异无统计学意义(P>0.05)外,硫酸镁组、糖苷生物碱组及马铃薯泥组t-PA及PAI-1表达均有所减弱(P<0.05)。结论:马铃薯提取物糖苷生物碱可通过下调t-PA、PAI-1水平改善化疗性静脉炎的病理损伤程度,效果优于马铃薯汁及马铃薯泥组,提示糖苷生物碱是马铃薯防治静脉炎的有效成分之一。
Abstract:
1 Gansu University of Chinese Medicine, Gansu Provincial Key Laboratory for Molecular Medicine and TCM Prevention and Treatment for Major Diseases in Colleges and University;2 Key Laboratory for Transfer of Dunhuang Medicine at the Provincial and Ministerial Level;3 Emergency Department, Gansu Provincial Hospital Objective: To study the effects of potato extract saccharide alkaloids, potato juice and mashed potato on the expressions of PAI-1 and t-PA, and the temperature of rabbit ear injection site of rabbit models with chemotherapy-induced phlebitis(CIP). Methods: Sixty Janpanese rabbits were radomized into blank control group, the model group, magnesium sulfate group, saccharide alkaloids group, potato juice group and mashed potato group, CIP rabbit models were established through injecting vinorelbine tartrate injection (VIN) via auricular vein, after establishing the models, the corresponding herbs were smeared on to rabbits puncture places, the temperature of rabbit ear injection site were measured at the corresponding time points, immunohistochemical method was adopted to measure the contents of t-PA and PAI-1. Results: 1) after modeling, rabbit ear temperature rose compared with before modeling (P<0.01), rabbit ear temperature lowered when saccharide alkaloids at the second day after treating, magnesium sulfate group, mashed potato group and saccharide alkaloids group at the third day after treating and the same groups after modeling were compared with model control group at the same time points, and the difference had statistical meaning(P<0.05); 2)the expressions of t-PA and PAI-1: fewer expressions of t-PA and PAI-1 could be seen under the microscope in the rabbit ear tissue of blank control group, and they presented in the tan granules; strong positive expressions of t-PA and PAI-1 could be found in perivascular tissue and intravascular thrombus (P<0.01), when magnesium sulfate group, saccharide alkaloids group, potato juice group and mashed potato group were compared with the model group, the difference had no statistical meaning in the expressions of PAI-1 except potato juice group (P>0.05), the expressions of t-PA and PAI-1 weakened in other groups (P<0.05). Conclusion: Potato extract saccharide alkaloids could improve pathological damage degress of CIP by downregulating the expressions of t-PA and PAI-1, and its effects are superior to potato juice and mashed potato group, which suggests that saccharide alkaloids is one of active ingredients of potato in the prevention and treatment for phlebitis.
