摘要:
目的:基于代谢组学方法,分析过敏性鼻炎(allergic rhinits,AR)肺虚感寒证和肾阳亏虚证患者血清游离脂肪酸(FFA)谱的变化,探讨脂肪酸与中医证候的关系,为两种中医证型的科学辨证提供依据。方法:采用GC/MS技术检测过敏性鼻炎肺虚感寒证和肾阳亏虚证患者血清游离脂肪酸,利用SIMCA-P软件进行主成分分析(principal component analysis,PCA)及偏最小二乘法判别(PLSA-DA)分析,研究血清游离脂肪酸在两组间的差异。结果:AR肺虚感寒证、肾阳亏虚证患者游离脂肪酸(FFA)谱分离趋势良好。AR肺虚感寒证和肾阳亏虚证患者筛选得到了花生酸(C20:0)、二十一酸(C21:0)、榆树酸(C22:0)3个差异有统计学意义的血清游离脂肪酸。并对3个显著差异的脂肪酸进行了ROC曲线绘制,计算ROC曲线下的面积,得出C20:0曲线下的面积为O.778,C21:0曲线下的面积为O.764,C22:0曲线下的面积为O.792.结论:血清游离脂肪酸(FFA)谱的变化与过敏性鼻炎证候有一定的关系,花生酸(C20:0)、二十一酸(C21:0)、榆树酸(C22:0)可用于区分过敏性鼻炎肺虚感寒证和肾阳亏虚证患者潜在生物标志物。
Abstract:
Objective: To analyze the changes of serum free fatty acid (FFA) spectrum in allergic rhinits (AR) of lung deficiency and cold pattern and kidney-yang deficiency pattern based on metabolomics, and explore the relationship between fatty acid and TCM syndrome in order to provide the reference for scientific syndrome differentiation for two patterns. Methods: GC/MS technology was used to detect FFA of AR patients of lung deficiency and cold pattern and kidney-yang deficiency pattern, SIMCA-P software was utilized to perform PCA and PLSA-DA, and study the difference of FFA among different groups. Results: FFA spectrum in AR patients of lung deficiency and cold pattern and kidney-yang deficiency pattern demonstrated good separation trends. After screening, three kinds of FFAs including arachidic acid (C20:0), heneicosoic acid (C21:0) and behenic acid (C22:0) which were significantly different have been obtained. ROC curves of three different FFAs haven been drawn, the area under ROC cure was calculated, the area under C20:0 curve was 0.778, under C21:0 curve 0.764 and under C22:0 curve 0.792. Conclusion: The changes of FFA spectrum are related to the patterns of AR certainly, arachidic acid (C20:0), heneicosoic acid (C21:0) and behenic acid (C22:0) could be used as the potential biomarkers for differentiating the patients of lung deficiency and cold pattern and kidney-yang deficiency pattern.
