摘要:
目的:观察白头翁汤对放射性肠炎大鼠一般情况及直肠组织纤维化相关因子的影响。方法:将100只健康雌性SD大鼠随机分为正常组、造模组、Y-27632组及白头翁汤低、高浓度组,每组20只。完成大鼠放射性肠炎的造模。正常组不造模,正常饲养;造模组以生理盐水灌肠;Y-27632组以Rho激酶抑制剂(Y-27632)灌肠;白头翁汤低、高浓度组以相应剂量白头翁汤灌肠。测定大鼠体质量、HE染色观察大鼠直肠组织病理学改变;Masson染色观察大鼠直肠组织胶原纤维沉积和变化;免疫组化分别检测第3、7、14、28天时I型胶原蛋白(type Ⅰ collagen,CollagenI)、转化生长因子β1(transforming growth factor-β1,TGF-β1)、Smad3蛋白的表达。结果:入组时各组大鼠体质量差异无统计学意义(P>0.05),与正常组相比,其余4组大鼠随着时间发展体质量均下降,其中造模组体质量下降较Y-27632组及白头翁汤低、高浓度组更明显。HE染色显示:与造模组相比,Y-27632组及白头翁汤低、高浓度组的中性粒细胞浸润,黏膜水肿、充血等现象随着时间的推移,均比造模组好转。Masson染色显示:与造模组相比,Y-27632组及白头翁汤低、高浓度组的胶原纤维增生降低。与造模组相比,Y-27632组、白头翁汤低浓度组和白头翁汤高浓度组的CollagenI、TGF-β1、Smad3表达均下调(P<0.05)。结论:白头翁汤能改善大鼠肠黏膜的炎症、水肿、充血;减少胶原纤维增生;可能机制是它下调了纤维化相关因子CollagenI、TGF-β1、Smad3的表达。
Abstract:
Objective: To observe the influence of Baitouweng decoction on general conditions and rectal fibrosis-related factors of the rats with radiation enteritis (RE). Methods: All 100 healthy female SD rats were randomized into the normal group, the modeling group, Y-27632 group, the groups of low and high concentrations of Baitouweng decoction. The normal group accepted normal feeding, after finishing the modeling of RE rats, the modeling group accepted physiological saline enema; Y-27632 group Y-27632 enema; the groups of low and high concentrations of Baitouweng decoction the corresponding decoction enema. To detect body mass changes of the rats, HE staining was used to observe pathological changes of rectal tissue; Masson staining to survey collagen deposition and changes in rat rectal tissue; immunohistochemical method to detect the expressions of Collagen I, TGF-β1 and Smad3 at the 3rd, 7th, 14th and 28th day respectively. Results: There was no difference in body mass of the rats in different groups when joining the group (P>0.05), compared with the normal group, as time passed, body mass of the rats in other four groups reduced, among them, decreased body mass of the modeling group was more notable than that of Y-27632 group, the groups of low and high concentrations of Baitouweng decoction. HE staining showed: compared with the modeling group, the phenomena of neutrophil infiltration, mucosal edema, congestion and others in Y-27632 group, the groups of low and high concentrations of Baitouweng decoction have improved as time passed. Masson staining showed: compared with the modeling group, collagen hyperplasia reduced in Y-27632 group, the groups of low and high concentrations of Baitouweng decoction. Compared with the modeling group, the expressions of Collagen I, TGF-β1 and Smad3 down regulated in Y-27632 group, the groups of low and high concentrations of Baitouweng decoction (P<0.05). Conclusion: Baitouweng decoction could improve the inflammation, edema and congestion of intestinal mucosa, and reduce collagen hyperplasia; and the possible mechanism is that it could down regulate the expressions of Collagen I, TGF-β1 and Smad3.
