摘要:
目的:建立测定大鼠血浆中5-氟尿嘧啶(5-fluoro,5-FU)的反相高效液相色谱(HPLC)分析方法,考察鸦胆子油乳对大鼠血浆中5-FU药动学的影响。方法:将SD大鼠平均分为5-FU组和5-FU/鸦胆子油联用组,每组6只。5-FU组大鼠尾静脉注射5-FU,联用组大鼠尾静脉注射5-FU及鸦胆子油乳,采用HPLC法测定大鼠血浆5-FU浓度。采用DAS Ver2.0药动学软件处理血药浓度数据,用SPSS 20.0统计学软件对所得药动学数据进行统计学差异分析。结果:与5-FU组比较,5-FU鸦胆子油乳联用组,5-FU在大鼠体内的半衰期T1/2,表观分布容积(Vd)增加,清除率CL降低,两组其他药动学参数比较差异均无统计学意义(P>0.05)。结论:鸦胆子油乳能延长5-FU在大鼠体内的半衰期,促进5-FU由血液向组织中分布。
Abstract:
Objective: To establish a method of RP-HPLC of detecting 5-FU in the plasma of the rats, and investigate the effects of Yadanzi (brucea javanica) oil emulsion on internal pharmacokinetics of 5-FU in rats. Methods: All the rats were divided into 5-FU group and 5-FU/yadanzi oil emulsion group evenly, the rats in 5-FU group accepted caudal vein injection of 5-FU, and combination group caudal vein injection of 5-FU and Yadanzi oil emulsion, HPLC method was used to detect the drug concentrations after giving 5-FU. DAS ver2.0 pharmacokinetics software was adopted to manage the data of blood drug concentrations, SPSS20.0 statistical software was used to analyze statistical difference of pharmacokinetics data. Results: Combined with Yadanzi oil emulsion, half-time of 5-FU in the rats was T1/2, apparent volume of distribution increased notably, clearance rate (CL) reduced remarkably, no significant difference has been seen in other pharmacokinetic parameters between both groups(P>0.05). Conclusion: Yadanzi oil emulsion could prolong half-time of 5-FU in rat body and promote its distribution from blood to tissue.
