摘要:
目的:分析中药挥发油对盐酸巴马汀纳米柔性脂质体(nano flexible liposomes,PFL)药剂学性能产生的影响。方法:选取5种常见的中药挥发油作为研究试剂,分别将其包埋于PFL的脂质双分子层内,利用体外泄露曲线图评估中药挥发油对PFL药物泄露产生的影响,并对艾叶油(folinm artemisisa argri oil,FA)对PFL包封率、结构、粒径、表面电位和变形性产生的药剂学性能影响。结果:不同中药挥发油均可以有效降低水溶性巴马汀(palmatine chloride,PC)从PFL内水相中的泄露,桉叶油(folium eucalypti oil,FE)、茉莉油(arabian jasmine oil,AJ)和丁香油(syzygium aromaticum oil,SA)挥发油的药物泄露程度相似(P>0.05),但FE、AJ和SA对PC泄露的抑制程度均明显高于FA和连翘油(fructus forsythisa oil,FF)(P<0.05),且FA各个时间段的药物泄露程度均明显低于其他4种挥发油(P<0.05);FA包埋前后的PFL在包封率结构、粒径、表面电位和变形性各方面差异均无统计学意义(P>0.05)。结论:选择合适的中药挥发油对PFL进行包埋可以有效抑制内水相中药物的泄露,其中以FA的抑制效果最好,有利于增加药物的吸收。
Abstract:
Objective: To analyze the effects of herbal volatile oil on pharmaceutical properties of nano flexible liposomes (NFL) in palmatine hydrochloride. Methods: Five kinds of common herbal volatile oils were taken as the reagents, and they were embedded into lipid bilayer of NFL, external leakage curve was used to assess the effects of herbal volatile oil on drug leak of NFL, and explore the effects of FA on pharmaceutical properties of deformability, surface potential, particle size, the structure and embedding ratio of NFL. Results: Different herbal volatile oil could effectively decrease the leakage of hydrosoluble PC from internal water phase of NFL, drug leakage degrees of FE, AJ and SA volatile oils were similar (P>0.05), while the inhibitory degrees of FE, AJ and SA to PC were higher than these of FA and FF obviously (P<0.05), and drug leakage degrees of FA at different time periods were lower than these of other four kinds of volatile oils notably (P<0.05); no significant difference has been found in the aspects of deformability, surface potential, particle size, the structure and embedding ratio before and after embedding of FA (P>0.05). Conclusion: Choosing appropriate herbal volatile oil to embed NFL could effectively inhibit the drug leakage in internal water phase, among them, the inhibitory effects of FA the best, and it is helpful to enhance the absorption ability of the herbs.
