摘要:
目的:探讨香茅草黄酮提取物对高血压大鼠降压作用及其机制。方法:用N’-硝基-L-精氨酸(L-NNA)给药4周复制高血压大鼠模型,将50只雄性高血压大鼠随机分为5组:模型组,香茅草黄酮提取物低剂量组(0.85%)、中剂量组(1.65%)、高剂量组(3.35%),卡托普利组(1%),每组10只;另外选取正常10只大鼠作为空白组。每日给予各组大鼠2 mL相应药物灌胃,空白组和模型组给予等量生理盐水。采用无创尾动脉测压法,每周测定大鼠收缩压(systolic pressure,SP)、舒张压(diastolic pressure,DP)1次,共4周。4周后,自腹主动脉取血,检测大鼠血清中超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malonaldehyde,MDA)、活性氧(reactive oxygen species,ROS)、一氧化氮(nitric oxide,NO)及内皮素(endothelin-1,ET-1)的含量。结果:给药4周后,与模型组相比,香茅草黄酮提取物低、中、高剂量组,卡托普利组能显著降低大鼠收缩压、舒张压、平均动脉压(P<0.05),血清中SOD的含量显著高于模型组(P<0.05),MDA、ROS的含量均显著低于模型组(P<0.05),香茅草提取物中、高剂量组以及卡托普利组大鼠血清中NO水平高于模型组,ET-1水平低于模型组,差异有统计学意义(P<0.05)。结论:香茅草提取物对L-NNA诱导的高血压大鼠血压有降压作用,其降压作用机制可能与香茅草提取物的抗氧化应激作用改善内皮功能有关。
Abstract:
Objective: To discuss the effects of flavonoid extract from Xiangmaocao (Cymbopogon citratus.) on lowering blood pressure of rat model with hypertension and its mechanism. Methods: L-NNA was given for four weeks to prepare hypertension rat models, fifty male hypertesion rats were randomized into five groups: the model group, low (0.85%), moderate (1.65%) and high (3.35%) doses groups of flavonoid extract from Xiangmaocao and the group of 1% captopril, ten rats each group, in addition, another ten normal rats were chosen as the blank group. Every day, the rats in different groups were drenched with the corresponding medicine, 2 mL, the blank group and the model group were given the same amount of normal saline. Noninvasive tail artery manometry was used to measure SP and DP each week, four weeks altogether. The contents of ET-1, NO, ROS, MDA and SOD in the serum were detected in four weeks after blood was drawn from abdominal aorta. Results: After medicating for four weeks, compared with the model group, low, moderate and high doses groups of flavonoid extract from Xiangmaocao and captopril group could notably lower SP, DP and MAP (P<0.05), while higher than the model group in the contents of SOD (P<0.05), lower than the model group in the contents of MDA and ROS (P<0.05), moderate and high dose groups of flavonoid extract from Xiangmaocao and captopril group were higher than the model group in the levels of NO, lower than the model group in the levels of ET-1, and the difference was statistically significant (P<0.05). Conclusion: Flavonoid extract from Xiangmaocao could lower blood pressure of rat model with hypertension induced by L-NNA, and its mechanism of lowering blood pressure might be related to that the extract from Xiangmaocao could fight against oxidative stress and improve endothelial function.
