摘要:
目的:探讨龙胆抗炎镇痛及治疗肝炎的药效物质基础及作用机制。方法:借助中药整合药理学平台,结合中医药大数据,按照“理法-方药-成分-靶标-通路-活性”关联性研究,探讨其抗炎镇痛及治疗肝炎的药效物质基础及作用机制。结果:共测得龙胆活性成分16个,分别作用于1个共同靶标,11个直接靶标和30条主要通路,其药效物质基础主要为环烯醚萜、黄酮、生物碱和三萜等成分。其抗炎镇痛及治疗肝炎作用涉及的1个共同靶标为:成纤维细胞;11个直接靶标主要有:ENPP1基因、ATP合成酶亚基δ(ATP5D)、GNG2基因、PTGER1抗体、白细胞介素1β(interleukin-1β,IL-1β)、Toll样受体7(toll-like receptor-7,TLR7)、RHO蛋白等;其抗炎镇痛及治疗肝炎作用涉及内分泌系统、循环系统、神经系统、免疫系统、Toll样受体信号通路、趋化因子信号通路等。结论:龙胆的主要活性成分,可针对多靶点、多通路发挥抗炎镇痛及治疗肝炎的作用。
Abstract:
Objective: To discuss pharmacodynamic material basis and the mechanism of Longdan (Gentiana scabra Bunge.) fighting against inflammation, easing pain, and treating hepatitis. Methods: With integrated pharmacology platform of TCM, combined with TCM big data, pharmacodynamic material basis and the mechanism of Longdan (Gentiana scabra Bunge.) fighting against inflammation, easing pain, and treating hepatitis were explored according to association study of "principle and method-precription and herbs-ingredients-target-pathwayactivity". Results: Altogether 16 kinds of active ingredients were obtained, respectively working on one common target, 11 direct targets and 30 main pathways, and its pharmacodynamic material basis were Iridoids, flavone, alkaloid, triterpenoids and other ingredients mainly. One common target involved in its anti inflammatory and analgesic effects and therapeutic effects for hepatitis was fibroblasts, 11 direct targets were ENPP1 gene, ATP5D, GNG2 gene, PTGER1 antibody (inhibitory effect of PGEG1 deficiency on kidney injury in diabetic mice), IL-1β, TLR7, RHO protein and others; its therapeutic effects involved endocrine system, circulatory system, nervous system, immune system, Toll like receptor signalling pathway, chemokine signaling pathway and others. Conclusion: Main active ingredients of Longdan could develop the effects of fighting against infammation, easing pain and treating hepatitis by aiming at many targets and many pathways.
