摘要:
目的探讨金刚藤与小果倒地铃合剂对链脲佐菌素(streptozotocin,STZ)诱导的2型糖尿病小鼠模型胰腺β细胞的抗炎及抗氧化作用。 方法采用40 mg/kg腹腔注射STZ诱导8周龄KM雄性小鼠糖尿病模型共15只,然后将模型小鼠随机分为模型组、合剂组、二甲双胍组各5只;另选相同来源的KM雄性小鼠10只作为对照组(分为空白对照组和合剂对照组);模型组每日给予高糖高脂饲料;合剂组饲喂高糖高脂饲料,灌胃合剂13.5 g/kg;二甲双胍组喂高糖高脂饲料,灌胃二甲双胍片溶液;空白对照组常规饲养,灌胃生理盐水;合剂对照组常规饲养,灌胃合剂13.5 g/kg;各组连续饲养8周。检测给药前、末次给药后小鼠空腹血浆血糖(fasting blood glucose,FBG)水平,ELISA法测定小鼠血浆中胰岛素分泌水平及胰腺组织中肿瘤坏死因子α(tumor necrosis factor α,TNF-α)、一氧化氮(nitric oxide,NO)、活性氧(reactive oxygen species,ROS)及核转录因子κB(nuclear factor-κB,NF-κB)水平;采用Western blot法测定胰腺组织中兔抗Tribble同源蛋白3(TRIB3)、兔抗小鼠C/EBP同源蛋白(CHOP)表达水平;采用多元线性回归方程分析TRIB3、CHOP表达水平与TNF-α、NO、ROS、NF-kB的相关性。 结果1)末次给药后,与空白对照组及合剂对照组比较,模型组、合剂组、二甲双胍组空腹血糖水平升高,血清胰岛素水平升高(P<0.05);与模型组比较,合剂组FBG水平降低,血清胰岛素水平升高(P<0.05);2)与空白对照组及合剂对照组比较,模型组、合剂组、二甲双胍组TNF-α、NO、ROS、NF-κB水平及TRIB3、CHOP表达水平升高(P<0.05);合剂组TNF-α、NO、ROS、NF-κB及TRIB3、CHOP水平低于模型组(P<0.05);3)TRIB3、CHOP表达与TNF-α、NO、ROS、NF-κB水平呈显著正相关(P<0.05)。 结论金刚藤与小果倒地铃合剂可能具有通过抑制氧化和内质网应激以及激活其抗氧化、抗炎和抗凋亡作用来保护胰腺β细胞并改善其功能的潜力。
Abstract:
ObjectiveTo explore anti-inflammatory and antioxidant effects of Jingangteng (smilax china) and Xiaoguo Daodiling (small-fruited balloon vine) mixture on pancreatic β cells in STZ-induced diabetic mice. MethodsAll 15 eight-week-old KM male mice were induced into DM models by intraperitoneal injection of STZ, 40 mg/kg, and then randomly divided into the model group, the mixture group, and metformin group, five mice each group; another ten KM male mice from the same source were used as the control group (divided into blank control group and mixture control group); the model group were given high-sugar and high-fat diet each day; the mixture group were fed with high-sugar and high-fat diet, and the mixture by gavage, 13.5 g/kg; metformin group high-sugar and high-fat diet, and metformin tablet solution by gavage; blank control group accepted conventional feeding and physiological saline by gavage; mixture control group conventional feeding and the mixture by gavage, 13.5 g/kg; different groups were raised for eight weeks consecutively. To detect the levels of FBG of the rats before medication and after the last medication, ELISA method was adopted to measure insulin secretion levels in mice plasma, the levels of TNF-α, NO, ROS and NF-κB in pancreatic tissue; Western blot method was used to detect the expressions of TRIB3 and CHOP in pancreatic tissue; multiple linear regression equation was used to analyze the connections between the expressions of TRIB3 and CHOP, and the levels of TNF-α, NO, ROS and NF-κB. Results1) after the last administration, compared with blank control group and mixture control group, the levels of FBG rose and the levels of serum insulin decreased in the model group, the mixture group and metformin group (P<0.05); compared with the model group, the levels of FBG lowered, and the levels of plasma insulin rose in the mixture group (P<0.05); 2) compared with blank control group and mixture control group, the expressions of TRIB3 and CHOP, and the levels of TNF-α, NO, ROS and NF-κB rose in the model group, the mixture group and metformin group (P<0.05); the expressions of TRIB3 and CHOP, and the levels of TNF-α, NO, ROS and NF-κB of the mixture group were lower than these of the model group (P<0.05); 3)the expressions of TRIB3 and CHOP were positively related to the levels of TNF-α, NO, ROS and NF-κB (P<0.05). ConclusionJingangteng and Xiaoguo Daodiling mixture could protect pancreatic β cells and improve its functional potential which might be realized through inhibiting oxidative stress and endoplasmic reticulum stress and activating its antioxidant, anti-inflammatory and anti-apoptotic effects.
