摘要:
目的观察青藤碱联合脉冲射频疗法对脊髓神经结扎大鼠(spinal nerve ligation,SNL)疼痛缓解的效果,探讨Wnt/β-catenin信号通路缓解SNL大鼠外周神经痛的作用及机制。 方法构建SNL持续性疼痛大鼠模型,设置假手术组、模型组、青藤碱组、脉冲射频组及联合治疗组,每组27只。观察各组大鼠治疗前后机械缩足反应阈值(mechanical withdraw threshold,MWT)、无翅型MMTV整合位点家族成员3α(wingless-related MMTV integration site-3α,Wnt-3α)、β-连环蛋白(β-catenin)、肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)和白细胞介素1β(interleukin-1 beta,IL-1β),伤害性信息与即刻早期基因c-fos的表达水平。 结果术后MWT均先降低后升高,且联合治疗组下降最慢而上升最快;Wnt-3α、β-catenin、TNF-α、IL-1β及c-fos的表达量随手术后时间的增长而降低,其中联合治疗组表达量降低的最为显著,与其他组比较差异有统计学意义(P<0.05)。 结论青藤碱联合脉冲射频疗法对大鼠SNL疼痛的治疗效果显著优于青藤碱及脉冲射频疗法单独治疗时的效果,其机制可能与抑制Wnt/β-catenin信号通路,降低背根神经节TNF-α、IL-1β、和c-fos的表达相关。
Abstract:
ObjectiveTo observe the effects of sinomenine and pulsed radiofrequency ablation on relieving pain in SNL rats, and explore the mechanism and the effects of Wnt/β-catenin signaling pathway in alleviating peripheral neuropathic pain in SNL rats. MethodsAfter establishing SNL rat models with persistent pain, they were allocated to sham operation group, the model group, sinomenine group, pulsed radiofrequency ablation group and combined treatment group, 27 rats in each group. To observe MWT, the expressions of Wnt-3α, β-catenin, TNF-α, IL- 1β and nociceptive information and immediate early gene c-fos of the rats in different groups before and after treating. ResultsMWT decreased first and then increased after the operation, and it presented the slowest decline and the fastest rise in the combined treatment group; the expressions of Wnt-3α, β-catenin, TNF-α, IL- 1β and c-fos lowered with the increase of postoperative time, among them, these expressions in the combined treatment group decreased the most significantly, the difference showed statistical meaning when it was compared with the other groups (P<0.05). ConclusionSinomenine and pulsed radiofrequency ablation are significantly better than single therapy in relieving pain in SNL rats, and its mechanism might be related to inhibiting Wnt/β-catenin signaling pathway, decrease the expressions of TNF-α, IL- 1β and c-fos of dorsal root ganglion.
